The method comprises the following steps of S1, inoculating activated green alga cells into a nitrogen-free sugar-containing culture medium, dividing the activated green alga cells into n parts, then adding the chemical inducer with a concentration gradient for induction culture to sequentially obtain inducer group green alga cells, and meanwhile, obtaining control group green alga cells by taking the condition that no chemical inducer is added as a control; S2, measuring the average fluorescence intensity of chlorophyll in the inducer group green alga cells and the control group green alga cells by adopting a flow cytometer; and S3, judging whether the chemical inducer can increase the oil yield in the green alga cells or not according to the average fluorescence intensity of the chlorophyll in the inducer group green alga cells and the control group green algacells, and accordingly screening out the optimal induction concentration.
