Problems solved by technology

Many researchers prefer to build models by inoculating tumor tissue blocks. This method also has certain shortcomings. One is that the composition of tumor tissue blocks is complex, not entirely a single tumor cell, and often mixed with other tissues
The second is the embedding of intrahepatic tumors, large wounds, heavy bleeding, high surgical requirements, and difficult to control the amount of transplantation
[0004] Various animal models of liver cancer that currently exist have certain defects and cannot meet the needs of the current biomedical field. Therefore, it is necessary to seek a new liver cancer modeling method

Embodiment Construction

[0027] In order to make the technical problems, technical solutions and advantages to be solved by the present invention clearer, the following will describe in detail in conjunction with specific embodiments.

[0028] A method for preparing an animal model of liver cancer, comprising the following specific steps:

[0029] Select 4-week-old ICR mice, observe and feed them for 2 weeks; HepG2 cells are cultured in DMEM medium supplemented with 10% fetal bovine serum, subcultured and expanded, washed 2-3 times with PBS, resuspended in 400 μL PBS, and used Inject the syringe into the peritoneal cavity of ICR mice;

[0030] After the mice developed obvious ascites, draw out 2 mL of ascites, add it to a centrifuge tube, centrifuge, and discard the supernatant; add 5 mL of PBS solution to resuspend, rinse, centrifuge for 5 minutes, and discard the supernatant;

[0031] Repeat the washing with PBS solution for 2-3 times in the same way as in step (2);

[0032] After discard...

Abstract

The invention provides a preparation method of a liver cancer animal model. The method comprises the following specific steps that (1), a 4-week-old ICR mouse is selected, observing and breeding are conducted for 2 weeks, HepG2 cells are subjected to culturing, passage and amplification in a DMEM culture medium mixed with 10% fetal bovine serum and then injected into the abdominal cavity of the ICR mouse through a syringe; (2), 2 mL of ascites is extracted after the mouse has obvious ascites, and added into a centrifuge tube, centrifugation is conducted, supernatant is discarded, 5 mL of a PBSsolution is added for resuspending and purging, then centrifugation is conducted for 5 minutes, and supernatant is discarded; (3), washing is conducted repeatedly 2-3 times according to the same method in step (2) through the PBS solution; (4), 5 mL of a PBS solution is added in sediment obtained after the supernatant is discarded in step (3) to blow away the cells, centrifugation is conducted for 3 minutes, erythrocyte of the upper layers of the supernatant and the sediment are taken out through a precise pipettor, and white sediment on the lower layer is retained. The effect of the preparedprimary liver cancer mouse model is greatly superior to an existing primary liver cancer mouse model building method at present, and compared with an existing model, the model is more realistic, practical, convenient and economical.

Description

technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to a method for preparing an animal model of liver cancer. Background technique [0002] Primary liver cancer is a notorious cancer in the world. With its late detection and high mortality rate, it ranks in the forefront of cancer mortality, accounting for 54% of new and dead patients worldwide every year. Due to the eating habits and susceptibility of the population in our country, there is a high incidence of hepatitis among Chinese people. If the treatment is not timely, it will often induce liver cancer. The heterogeneity of the population, the general resistance to clinical research, and the limitations of scientific ethics review have hindered clinical liver cancer research. Therefore, the majority of scientific workers are committed to developing excellent animal models to replace human liver cancer research, and achieved many results. However, the existing animal m...
