Embodiment 1

[0036] Example 1. Fucoidin improves glomerular morphology and cell matrix accumulation in rat kidney tissue

[0037] Renal fibrosis is typically characterized by abnormal accumulation of extracellular matrix in the glomerular mesangial region. In this example, rats were used as a model, and the effect of fucoidin on glomerulus morphology and extracellular matrix accumulation in rat kidney tissue was studied by HE staining.

[0038] experimental method:

[0039] SD rats (Sprague Dawley rats, purchased from the Animal Experiment Center of Sun Yat-Sen University); streptozotocin (Sigma, USA); fucoidin (Chengdu Purifa Biotechnology Co., Ltd.); Dye, purchased from Wuhan Sewell Biotechnology Co., Ltd.); Chloral hydrate (Tianjin Baishi Chemical Reagent Company); olive oil (Shanghai Linen Technology Development Co., Ltd., analytically pure grade)

[0040] Diabetic nephropathy is a disease in which hyperglycemia induces renal fibrosis. This field usually adopts freshly configured st...

Embodiment 2

[0045] Example 2: Fucoidin significantly reduces the protein content of FN and ColⅣ in rat kidney tissue

[0046] The typical specific feature of renal fibrosis is abnormal accumulation of extracellular matrix, and the main components of extracellular matrix are FN and ColⅣ. In this experiment, the effect of fucoidin on the expression levels of FN and ColⅣ in rat kidney tissue was determined by Western blotting. To reflect the effect of fucoidan on the accumulation of extracellular matrix in the kidney.

[0047] experimental method:

[0048] Using the rat kidney isolated in Example 1, take 10 mg of kidney tissue each, add 100 μl RIPA lysate (containing 1% protease inhibitor) for homogenization at 4°C, collect the tissue homogenate into a centrifuge tube, and centrifuge at 12000rpm at 4°C After 30 minutes, the protein supernatant was divided into 1.5ml centrifuge tubes. After BCA protein quantification operation, immunoblotting was carried out to detect the protein content of...

Embodiment 3

[0051] Example 3. Fucoidin significantly reduces the expression of FN and ColⅣ in rat glomerular mesangial cells

[0052] Glomerular mesangial cells are intrinsic cells in the glomerulus. These cells can produce extracellular matrix proteins, secrete cytokines, phagocytize and remove foreign bodies, regulate the flow in glomerular capillaries, and maintain the tissue structure and Physiological functions play an important role. In this example, glomerular mesangial cells were treated with high sugar to simulate the process of renal fibrosis caused by high blood sugar in diabetic nephropathy, and the effects of high sugar and fucoidin on the expression of FN and ColⅣ proteins in glomerular mesangial cells were studied .

[0053] experimental method:

[0054] Rat glomerular mesangial cells (hereinafter referred to as cells) were donated by the research group of Professor Huang Heqing from the Laboratory of Pharmacology and Toxicology, School of Pharmacy, Sun Yat-sen University...

Abstract

The present invention provides the use of fucoidin in preparing a medicine for treating and / or preventing renal fibrosis disease. The embodiment of the invention proves that the fucoidin has the following functions: 1) reduces abnormal accumulation of extracellular matrix in renal glomerular mesangial region by reducing the protein expression level of FN and Col IV which are abnormally increasedin glomerular mesangial cells; and 2) obviously reduces the oxidative stress level of glomerular mesangial cells and the expression of inflammatory factors. Through the functions, the fucoidin can effectively reduce the renal function damage caused by renal fibrosis so as to achieve the effect of treating and / or preventing the renal fibrosis diseases, and therefore, the fucoidin can be developedas a potential medicament for treating and / or preventing the renal fibrosis diseases.

Description

technical field [0001] The invention relates to the technical field of new uses of fucoidin, in particular to the use of fucoidin in the preparation of medicines for treating and / or preventing kidney fibrosis. Background technique [0002] Renal fibrosis is a common pathway through which chronic kidney disease eventually leads to end-stage renal failure. It is a gradual process in which the function of the kidney changes from healthy to damaged, then damaged, and finally lost. The kidney is stimulated by various pathogenic factors such as trauma, infection, inflammation, blood circulation disorder, and immune response, and its intrinsic cells are damaged. In the later stage of development, a large amount of collagen deposits and accumulates, causing the renal parenchyma to gradually harden and form scars, until the kidney is damaged. Complete loss of organ function. The process of intrinsic cell fibrosis and sclerosis in the kidney is also the process of renal fibrosis. Re...
