Embodiment 2

[0043] Genetic engineering application of embodiment 2 gene GmMYB32

[0044] 1) Cloning of soybean MYB32 transcription factor encoding gene GmMYB32

[0045] Using the root total RNA of soybean (Glycine max) phosphorus-sensitive varieties Silver and Honeycomb Bean 73 as templates, the first strand of cDNA was synthesized by reverse transcription, and PCR amplification was performed. The primer sequences are shown in SEQ ID NO.3 and SEQ ID NO. 4. The PCR program is as follows: pre-denaturation at 95°C for 3 minutes, denaturation at 95°C for 15 seconds, annealing at 60°C for 15 seconds, extension at 72°C for 1 minute, a total of 35 cycles, and a final incubation at 72°C for 5 minutes, followed by constant temperature at 12°C. The product was cloned into the pUC19-TVector vector, and after sequencing, the CDS sequence of the soybean GmMYB32 gene with a length of 1017 bp and a complete coding region was obtained, wherein the sequence of the coding region is shown in SEQ ID NO.1;

Abstract

The invention discloses an application of a soybean MYB32 transcription factor encoding gene GmMYB32. The nucleotide sequence of the soybean MYB32 transcription factor encoding gene GmMYB32 is represented by SEQ ID NO.1. A constructed plant overexpression vector pMDC83-GmMYB32 is heterologously expressed in wild Arabidopsis thaliana, and it is found that the overexpressing plant grows strong underlow-phosphorus conditions, the biomass is increased, the length of a main root and the fresh weight of an overground part of the overexpressing plant are significantly higher than those of the wild type, so the gene can be introduced to plants as a target gene, and the growth vigor of the plants can be enhanced to reverse the phosphorus deficiency symptoms of the plants and improve the low phosphorus tolerance of transgenic plants. The soybean MYB32 transcription factor encoding gene GmMYB32 can enhance the growth vigor of the plants and reverse the phosphorus deficiency symptoms of the plants by genetic engineering, so the low phosphorus tolerance of the transgenic plants is improved.

Description

technical field [0001] The invention relates to the application of soybean MYB32 transcription factor encoding gene GmMYB32, which belongs to the field of genetic engineering, and specifically relates to the enhancement of plant growth and biomass by the MYB32 transcription factor encoding gene derived from soybean, thereby enhancing the low phosphorus tolerance and yield of transgenic plants aspects of application. Background technique [0002] Phosphorus is one of the three essential elements for plants. Insufficient phosphorus supply will have adverse effects on plant growth and metabolism. The actual phosphorus concentration in the soil is far from meeting the needs of plants, and phosphorus fertilizers are usually required to supplement the phosphorus supply. However, the utilization rate of phosphorus fertilizer by crops is seriously insufficient. At present, the mechanism of plant regulation of phosphorus is still not fully resolved. Studying the function of genes ...
