Problems solved by technology

These methods can effectively suppress the browning phenomenon of licorice suspension cells in short-term cultivation (within 10 days), but cannot effectively inhibit the browning phenomenon of licorice suspension cells in long-term cultivation (12-20 days).
Replacing the medium and supplementing the anti-browning agent during the cultivation process will increase the pollution rate of the operation and increase the workload, which is not conducive to the scale-up cultivation
When the adsorbent is added to the culture medium, it will adsorb the hormone in the culture system, resulting in a decrease in the concentration of the hormone in the medium. At the same time, the anti-browning effect of the adsorbent is not significant, and the accumulation of biomass (wet weight of cells) does not change significantly.

Abstract

The invention discloses a method for inhibiting browning in glycyrrhiza glabra suspension cell culture process for long time, and aims at inhibiting browning in the glycyrrhiza glabra suspension cellculture process for long time. The method is characterized in that an ascorbic acid derivate (calcium ascorbate) is adopted to replace ascorbic acid as a browning resisting agent, and meanwhile, lactose is screened to replace saccharose as a carbon source in the culture of glycyrrhiza glabra suspension cells. The result shows that the activity of polyphenol oxidase in the suspension cells can be inhibited for long time (20d) by the culture method, and the browning in the glycyrrhiza glabra suspension cell culture process can be inhibited; and meanwhile, the biomass accumulation of the suspension cells is obviously increased. Additionally, the method is simple to operate; the obtained glycyrrhiza glabra suspension cells are high in dispersing performance and good in growing state. An effective technical mean is provided for amplification culture of glycyrrhiza glabra cells.

Description

technical field [0001] The invention relates to a method for culturing suspension cells of Glycyrrhizae glabra, in particular to a method capable of suppressing the browning phenomenon in the cultivation process of suspension cells of Glycyrrhiza glabra for a long time, and cultivating the suspension cells of Glycyrrhiza glabra. The invention belongs to the technical field of cell culture. Background technique [0002] Licorice (Glyrrhlza uralensis Fisch) belongs to the leguminosae plant and Papilionaceae subfamily, and is mainly distributed in the northeast, north and northwest of China. In China, there are Glycyrrhiza uralensis Fisch., Glycyrrhiza inflate Bat., Glycyrrhiza glabra L., Glycyrrhiza aspera Pall., Glycyrrhiza squamulosa Franch. , Glycyrrhiza inflata Batal., Glycyrrhiza pallidiflora Maxim., Glycyrrhiza yunnanensis Chengf. and Glycyrrhiza eglandulosa X.Y.Li. There are more than 400 known compounds in licorice, among which triterpenoid saponins and flavonoids ar...
