Abstract

Fresh euonymus bungeanus pollen is collected from a well-developed mother tree and then placed into 1.8-ml frozen preservation tube, the frozen preservation tube is then placed in a drying vessel containing a silica gel desiccant and dried for 10 hours to keep the pollen water content at 4%-4.5%; the frozen preservation tube with pollen is put into a liquid nitrogen tank at subzero 196 DEG C for preservation, and then defrosting is performed at the room temperature for 30 min; a culture solution formula adopted for germination of the defrosted pollen comprises sucrose with concentration of 100 g / L and boric acid with concentration of 150 mg / L. The method is easy to operate, safe and stable, occupied space is small, the preservation cost is low, the preservation time of pollen is greatly prolonged, long-term storage and exchange of pollen become possible, and the pollen after being preserved at the ultra-low temperature can preserve genetic integrity and stability.
