The invention discloses a method for rapid detection of the single nucleotide polymorphism of a sheep NELF gene by using PCR-SSCP and application thereof. According to the invention, the whole genome DNAs of a to-be-detected sheep are used as a template; a primer pair P is used as primers; PCR amplification is carried out so as to obtain a sheep NELF gene fragment; a PCR product undergoes denaturation so as to form single-stranded DNAs; and the single-stranded DNAs undergo electrophoresis in neutral polyacrylamide gel so as to produce different electrophoretic bands. The single nucleotide polymorphism of the site 454 of the sheep NELF gene is identified according to SSCP typing results. The method can rapidly detect the single nucleotide polymorphism of the sheep NELF gene and lays a foundation for establishment of a relationship between the single nucleotide polymorphism of the sheep NELF gene and reproductive performance, so basic data is provided for marker assisted selection of reproductive traits of Chinese sheep and rapid building of a sheep population with excellent genetic resources is facilitated.
