The invention aims to overcome the defect of the field of the present detection method for 4-EP and 4-EG in red wine and provides a method for simultaneous determination of the content of 4-EP and 4-EG in red wine by researching the optimal headspace solid-phase micro-extraction condition and the optimal gas chromatography-mass spectrum separation condition. The method mainly comprises the following steps: preparing an internal standard substance; performing headspace solid-phase micro-extraction; determining a standard curve; and performing gas chromatography-mass spectrum detection. The content of 4-EP and 4-EG in a sample can be calculated according to the obtained standard curve. Through verification, the method has the advantages of high speed, accuracy, convenience, higher precision and higher recovery rate.
