The method comprises a fresh semen collecting, disinfecting and diluting process and an induced ovulation and timed semen deposition process; firstly, collected seminal fluid is disinfected and diluted; then, a donkey can ovulate in the concentrated time by using a hormone injection method; the atresia probability in the follicular development process is reduced; disinfected and diluted fresh semens are subjected to artificial semen deposition within 10 hours before the ovulation and within 6 hours after the ovulation, and the semen deposition dose is 15mL; donkey semens and donkey ova are enabled to meet to complete the insemination process in the time period with insemination capability; and the donkey conception is realized.
