Abstract

The method disclosed by the invention comprises the following steps of scissoring an immature green fertile spike on the top end of the fertile spike of osmunda japonica; sterilizing an explant, cutting the fertile spike of osmunda japonica into sections, and inoculating the sections in an agar culture medium, and culturing; transferring germinal young prothallus in a prothallus proliferation agar culture medium at 23-26 DEG C with the illumination of 12h / d and the intensity of 2300-2500lx; taking out proliferated prothallus, putting the proliferated prothallus in a sterile triangular flask containing a liquid culture medium, carrying out spore culture in a culture medium containing 0.1mg / L of 1 / 2MS+6-BA and 0.01 mg / L of NAA, oscillating on a shaking table for 4-10min, respectively inoculating proper amount of prothallus in a culture bottle containing a filter paper bridge or cotton, and pouring into an MS liquid culture medium and culturing; and transferring germinal sporophore in the culture bottle containing the filter paper bridge or the cotton, and pouring into a sporophore proliferation liquid culture medium, and culturing.
