[0012] Take 10 pieces (1500±15g) of fresh pork liver samples after slaughter, and place them in an artificial climate box with a constant temperature of 25°C, a constant humidity of 70%, and 12 hours of daylight to allow them to autolyze and rot.

[0013] According to the sampling "six determination" method to standardize the sampling: 1. Timing: according to the time points of 2h, 12h, 18h, 24h, 30h, 36h, 48h, 60h, 72h, 84h, 96h, 108h, 120h, 132h, 144h, 156h and 168h Collect materials in groups; 2. Positioning: collect materials from the thickest part of the interstitial connective tissue in the left outer, left inner, right inner, and right outer four lobes of the liver, away from the large blood vessels and their surroundings; 3. Quantification: intercept the same weight Liver tissue specimen 10g; 4. Shaping: the sampling of different samples is uniformly cut into the same rectangular size to ensure that its volume and surface area are basically equal; 5. Fixed ratio: prepar...

Abstract

The invention relates to a method for enhancing the mechanics property of a biological tissue and prolonging the detectable time of the mechanics property of the biological tissue by using a stationary liquid. According to the method, a biological tissue stationary liquid takes a fixation effect by means of the crosslinking of protein molecules to fix in-vitro biological tissues of different PMIs (Postmortem Intervals), so that autolytic enzymes and putrefactive bacteria in cells of a biological tissue can be immediately inactivated to stop the autolysis and putrefaction processes of a dead body or in-vitro tissue; and meanwhile, a tissue structure is rapidly fixed and the biomechanics property of the tissue structure is enhanced to a certain extent, so that the PMI can be better deduced by utilizing a sensitive biomechanics property detecting index, the detectable time of the mechanics property of the biological tissue can be prolonged to a certain extent, and the time limit for deducing the optimal window period of the PMI can be prolonged. The stationary liquid is used for sampling a solid from the in-vitro biological tissues according to a six-fixation method: 1, fixed time; 2, fixed position; 3, fixed quantity; 4, shaping; 5, definite proportion; and 6, timed time. The stationary liquid can be any one common stationary liquid such as formaldehyde and alcohol.

Description

technical field [0001] The invention relates to a method for enhancing the mechanical properties of biological tissue and prolonging its detectable time with a fixative. Background technique [0002] Postmortem Interval (PMI) refers to the time interval from the moment of death to the moment of examination. In the detection of criminal cases, PMI is of great significance to the judiciary of determining the time of the incident, identifying suspects, and delineating the scope of investigation. It also has certain significance for the handling of inheritance, insurance claims and other cases. Since the time of occurrence of many unnatural death cases is not clear, how to accurately infer PMI has always been an important topic in the actual identification of forensic medicine. [0003] The current method of applying biomechanical technology is quite mature and sensitive. It only needs a small, regular-shaped standardized specimen of one-dimensional, two-dimensional or three-di...
