Embodiment 1

[0028] 1. Experimental materials

[0029] The terminal buds of aseptic seedlings germinated from mature cone seeds of Pine masson in the same year were used as explants, and the cones were collected from the Pine masson seed garden established by the excellent provenance of Gupeng in Xincheng, Guangxi.

[0030] 2. Experimental method

[0031] (1) Sterile explant selection and aseptic treatment

[0032] The newly harvested cones are first dried in the sun, and after the scales of the cones are opened, the seeds are gently shaken out, and the seeds are packed in plastic bags and placed in a refrigerator at 4°C for later use. The seeds refrigerated for one week were taken out of the refrigerator, soaked in 0.5% potassium permanganate solution for 15 minutes, then rinsed with running water for 5 minutes, soaked in deionized water for 12 hours, sterilized with 0.1% mercury liter for 40 minutes, and rinsed with sterile water for 5 times , directly sowed in the sand substrate steri...

Abstract

The invention discloses a tufted-bud induction and subculture method for southern pine trees. The method comprises the following steps of: using an apical bud of an aseptic seedling with a hypocotyl which is 0.5cm long as an explant, performing induction culture for 20-26 days by an improved MS culture medium, 2.5-4.0 mg / L 6-BA, 0.05-0.1 mg / L NAA, 30,000 mg / L sugar crane, 3,500 mg / L agar and 500 mg / L casein hydrolysate, then performing cluster bud extending and mulitiplication culture by a culture medium comprising the improved MS culture medium, 0.25-0.4 mg / L NAA or 3,000-5,000 mg / L active carbon, 30,000 mg / L sugar crane and 3,500 mg / L agar and a culture medium comprising the improved MS culture medium, 1.0-3.0 mg / L 6-BA, 0-1.0 mg / L KT, 0-0.3 mg / L NAA, 30,000 mg / L sugar crane and 3,500 mg / L agar, wherein the proportions of components of the culture media are different; repeating the subculture by the culture mode for N times, and obtaining large-scale transgenerational buds. According to the method, the cluster bud induction rate is over 97 percent; the bud seedlings are high in extending rate and super-vital; the bud yield is high; the multiplication coefficient is more than 6.8; high-quality and high-efficiency southern pine seedling forestation can be supplied; and the method has high economical and social benefits.

Description

technical field [0001] The invention belongs to the technical field of plant propagation, and relates to tissue culture seedling raising technology, in particular to a method for inducing and subculturing buds of southern pine bushes. Background technique [0002] As a tree species with high comprehensive utilization value and broad promotion prospects, pine trees can not only be used to produce three-board, paper and chemical fiber industrial manufacturing, but also can be used to produce industrial raw materials such as rosin and turpentine. In recent years, with the intensification of contradictions between people's growing living needs and the shortage of timber resources and the gradual depletion of non-renewable resources such as petroleum, the sustainable development of ecology, economy and society has attracted more and more attention from the society. Based on the problems of ecological safety, resource security and social security in our country, it is necessary to...
