Problems solved by technology

Maintaining accuracy and repeatability during the sampling process is important in automated sampling systems, and the surface layer of biomass often obstructs or obstructs the sampling needle of the automated pipetting station, which causes problems related to repeatability, reliability, and process flow

Embodiment 1

[0278] Example 1. Sampling with the aid of an adapter from a microtiter plate with Aspergillus oryzae

[0279] MDU-2BP medium: 45g maltose-1H2O, 7g yeast extract, 12g KH per liter 2 PO 4 , 1g MgSO 4 -7H 2 O, 2g K 2 SO 4 , 5g urea, 1g NaCl, 0.5ml AMG trace metal solution, pH5.0.

[0280] The filamentous fungus Aspergillus oryzae was grown in standard SBS microtiter plates (96 wells) in MDU-2BP medium at 30 °C for 4 days (200 µl / well). Following incubation, fungal growth was observed on the surface of the growth medium in the wells, forming a dense mat of organic material on the surface of the medium in each well.

[0281] exist Figure 4 The prototype adapter shown in was placed on top of the microtiter plate and pushed down as far as possible. The adapter fingers push the biomass pad into the liquid in each well, and the liquid flows around the perimeter of the pad and into the hollow adapter finger, where sampling can take place without the biomass obstructing or clog...

Embodiment 2

[0284] Example 2. Biomass transfer from a microtiter plate with Aspergillus oryzae by means of an adapter

[0285] The adapters described in Example 1 were tested using Aspergillus oryzae grown in 96-well microtiter plates. Not only can the adapter be used as a biomass pusher to push the biomass to the bottom of the wells of the microplate, allowing sampling of the liquid supernatant, as shown above, it can also be used to transfer the biomass to a new microplate well. plate, e.g. for storage, leaving the spent growth medium or supernatant in the original plate for easy sampling and further assays.

[0286] according to Figure 5 Drawings in the prototype of the adapter according to the invention were fabricated by 3D printing (stereolithography); pictures of the resulting device are at Figure 18 shown in .

[0287] exist Figure 4 Our first prototype shown in was designed for use with an autopilot with a fixed (thin) tip - we wanted to use it with a disposable pipette ti...

Abstract

The present invention relates to adapters for microplates. The adapters allow sampling from a liquid growth medium in a microplate well after a microorganism has formed a dense layer of biomass on the surface of the growth medium. The adapters also allow easy transfer of a surface-grown microorganism from all wells of a microplate in one go, when inserted before the microorganism form the layer and then removed after incubation and layer-formation, which also enables easy sampling of the growth medium in the microplate afterwards.

Description

technical field [0001] The present invention relates to adapters for microplates that allow sampling from liquid growth medium in the wells of the microplate even when microorganisms have formed a dense layer of biomass on the surface of the growth medium. When fitted into a microplate, the adapter pushes down on the surface layer of biomass in each well, allowing liquid media to flow around and over the biomass layer, allowing samples to be drawn from the media in the wells, without penetrating the biomass layer. [0002] When the adapter is assembled into the microplate prior to the incubation of the plate and the microorganisms form a layer of biomass on the surface of the liquid growth medium within the hollow fingers of the adapter, the adapter can also absorb the bacteria from all the wells in the microplate at once. Biomass of surface-grown microorganisms is readily transferred. After incubation, the adapter is lifted from the microplate and liquid growth medium flows...
