[0064] In order to study the feasibility and accuracy of detecting specific HLA genotypes by the new detection method using B*1502 as an example, in the first phase of the study, healthy blood donors were recruited anonymously and 50 DNA samples were obtained (B* 1502 positive 25 and B*1502 negative 25) and 200 frozen blood samples (B*1502 positive 30 and B*1502 negative 170). All of these 250 samples were typed in a previous HLA genetic association study by high-resolution SBT as a control and were thus known to be HLA-B genotyped. SBT was performed according to the IHWG technical manual (8), and the resulting sequences were analyzed using the SBT tool (Genome Diagnostics, the Netherlands). Carrier status of B*1502 was detected directly in DNA samples by the novel LAMP method, and in frozen blood samples after heat treatment without DNA extraction.
