Problems solved by technology

[0004] Superovulation is a quantitative trait, which is mainly controlled by multiple genes in addition to being affected by the environment. The more related genes screened out, the better the accuracy of selection and the improvement of selection intensity. However, only the genes FSHR and Mutations in two genes of INHA are associated with

Embodiment 1

[0022] Genomic DNA was extracted from cow blood with a genomic DNA extraction kit (Tiangen Biochemical Technology Co., Ltd.).

[0023] Partial cloning of cow LHCGR gene:

[0024] (1) Primer design: using bovine genomic DNA (gene database ID in NCBI: 281900) as a template, use Primer 5.0 to design a pair of specific primers:

[0025] The forward primer is 5'-GCTCTACCTGCTGCTCAT-3',

[0026] The reverse primer is 5'-TAATTGCTGACACCCACA-3'.

[0027] (2) PCR reaction system and amplification conditions: The total PCR reaction system is 20 μl, of which the forward and reverse primers are each 10 pM, dNTP is 4 μM, 10×PCR buffer (Mg2+plus) 2 μl, 0.5 units of Taq DNA polymerase (Bao Biological Engineering Co., Ltd.), genomic DNA 50ng. The PCR reaction program was pre-denaturation at 95°C for 5 min, followed by 35 cycles, each cycle including denaturation at 95°C for 30 s, annealing at 57°C for 30 s, extension at 72°C for 30 s, and extension at 72°C for 5 min outside the cycle. Abstract

The invention relates to application of an LHCGR gene of a Chinese Holstein cow used as a molecular marker. The LHCGR gene of the Chinese Holstein cow is positioned at a Reference SNP Cluster Report: rs41256848 site; and when the site is homozygosized into 'G', the advantage of superovulation is shown. The LHCGR gene serving as the molecular marker is used for the assistant breeding of the Chinese Holstein cow, so that the generation interval can be shortened and the breeding process can be improved.

Description

technical field [0001] The invention relates to the application of a gene with a mutation site in livestock as a molecular marker, in particular to the application of the Chinese Holstein cow LHCGR gene as a molecular marker in breeding superovulated dairy cows. Background technique [0002] At present, the traditional selection methods in dairy cattle breeding are mostly based on the phenotypic record value of the individual, and the genetic advantage of the individual in a certain trait is evaluated by analyzing the phenotypic data of the individual itself and its relatives. Ke-Qiong Tang et al. (2010) and Wu-CaiYang et al. (2010) used the genes INHA and FSHR as molecular markers for superovulation traits in recent studies on molecular marker-assisted selection for superovulation traits in Chinese Holstein dairy cows. . [0003] Due to the traditional selection method, the data of superovulation can only be obtained after the individual grows to sexual maturity or later, ...
