Embodiment 1

[0021] In the present invention, the terminal buds of the lotus root variety Elian No. 5 are used as explants, and the test-tube lotus is formed through stem tip initiation, subculture multiplication, rooting, and induction. Including the following steps:

[0022] 1. Stem tip initiation: Take the cleaned terminal buds of the disease-free Elian No. 5 variety, soak them in 75% alcohol for 30 seconds, and wash them with 0.1% HgCl 2 Soak in the solution for 3 to 12 minutes, then rinse with sterile water for 3 times, each time for 3 to 5 minutes. After completing the above surface disinfection treatment procedure, peel off the outer leaf sheath with a scalpel, cut off a 0.3-0.6cm long shoot tip, and inoculate it in lotus root shoot tip differentiation medium 1 / 2MS+6-BA2.0mg / L+NAA0.5mg / L+3.0% sugar, at a temperature of 24-26°C and a light intensity of 2000-2500 lx, and light for 10 hours a day, buds or clusters of buds will appear after 30-45 days. The pH value of the lotus root ...

Embodiment 2

[0027] The terminal buds of the seed lotus variety Kongkong No.3 were used as explants, and the test-tube lotus root was formed after shoot apex initiation, subculture multiplication, rooting, and induction. Including the following steps:

[0028] 1. Shoot apex start: Take Space No. 3 as the test material, take the clean and disease-free terminal shoot of Space No. 2 Soak in the solution for 3 to 12 minutes, then rinse with sterile water for 3 times, each time for 3 to 5 minutes. The pH value of the lotus root shoot tip differentiation medium is 5.8-6.0.

Abstract

The invention relates to an inducing technology of a test tube lotus root, which comprises the steps of: using terminal buds or lateral buds of the lotus root as explants; carrying out the starting of stem tips, successive proliferation and rooting; finally culturing and inducing by using a solid-liquid culture base to form the test tube lotus root. The method for inducing the test tube lotus root comprises the following steps of: cutting the stem tips of the lotus root and inoculating the stem tips on a stem tip starting culture base for culturing so as to grow up to become buds or a cluster of buds; transferring the buds or the cluster of buds to a successive culture base and carrying out successive proliferation; segmenting the cluster of buds into single bud and transferring to a rooting culture base and inducing to root to form tube seedlings; and rooting and culturing for 30d, pouring the liquid culture base into a culture container and inducing to form the test tube lotus root. In the process, the culture temperature is between 24 and 26 DEG C, the illumination intensity is 2000-25001x and the illumination is carried out for 10h every day. Through the stem tip culture and quick reproduction of the lotus root, the test tube lotus root is formed by inducing so as to quicken the reproduction speed of the lotus root, improve the transplant survival rate and industrially produce.

Description

technical field [0001] The invention relates to the field of agricultural biotechnology, in particular to a test-tube lotus root induction technique. Background technique [0002] my country is one of the origin centers of lotus root. Lotus root is widely distributed and cultivated in my country, and it is the aquatic vegetable with the largest cultivated area in my country. It is very common in the Yellow River Basin and the areas south of it. With the adjustment of the rural industrial structure, the demand for lotus root species has increased sharply, but because lotus root is a crop mainly asexually propagated, the reproduction coefficient is low (1:10), and the reproduction speed of excellent new lotus root species is far from meeting the market demand. Moreover, conventional lotus root seeds have problems such as large amount of seeds, high transportation costs, large loss, and easy to carry diseases, which limit the development of lotus root industry to a certain ext...
