[0042]The ability to detect leukocyte esterase in accordance with the present invention was demonstrated. Initially, an HF12002 nitrocellulose membrane laminated to a support card (Millipore, Inc.) was provided as the chromatographic medium. To form a detection zone, Diazo Red DC (DR) was initially striped by hand onto the membrane. The Diazo Red DC was in the form of a solution containing water and approximately 15 wt. % dimethylformamide (“DMF”). The membrane was then dried at 37° C. for two hours. A reagent pad was then fabricated from a glass fiber pad having a length of 15 centimeters (Millipore Inc.).
