Embodiment 1

[0027] Example 1 Obtaining SNP Detection Fragment of Pig NT5C1A Gene and Establishment of Detection Method

[0028] 1. Extraction of pig genomic DNA

[0029] The test pig breeds of the present invention are Sedu black pig and French Large White pig, and the samples are all provided by the breeding pig farm of the Institute of Animal Husbandry and Veterinary Medicine, Hubei Academy of Agricultural Sciences. Pig genomic DNA was extracted using the blood genomic DNA extraction kit (article number: DP348) produced by Beijing Tiangen Biochemical Technology Co., Ltd., and the specific steps were referred to the kit instructions. The concentration and quality of the extracted DNA were tested and stored at -20°C for future use.

[0030] 2. Obtaining the SNP genetic marker detection fragment of pig NT5C1A gene

[0031] (1) PCR amplification

[0032] According to the genome sequence of porcine NT5C1A gene (GenBank ID: NC_010448), the primer pair of above-mentioned SEQ ID NO:1 sequenc...

Embodiment 2

[0045] Example 2 Detection of Polymorphism Distribution of Molecular Markers in Different Pig Groups

[0046] In this example, the polymorphism of the C / T site at 302bp of the pig NT5C1A gene SEQ ID NO: 1 was detected in the populations of Sedu black pigs and French large white pigs respectively, and the detection results are shown in Table 1.

[0047] Table 1 Distribution of c.302C / T polymorphism in exon 6 of NT5C1A gene in different pig groups

[0048]

[0049] From the results in Table 1, it can be seen that the c.302C / T locus of exon 6 of the NT5C1A gene has three genotypes in different pig populations, among which the CC genotype is dominant, and the C allele is the dominant allele , the proportion of C allele in local pig breed Sedu black pig is higher than that in foreign pig breed French line Large white pig.

Embodiment 3

[0050] Example 3 Association Analysis and Application of Molecular Markers and Pork Quality Traits

[0051] In order to determine whether the c.302C / T polymorphism of exon 6 of the NT5C1A gene is related to pork quality traits, 398 French large white pigs × Sedu black pigs F 2 The generation resource population was the test material, and the method established in Example 1 was used for polymorphism detection, and the SAS statistical software (SAS Institute Inc, Version 9.1) GLM program was used for single-marker analysis of variance to analyze the porcine NT5C1A gene exon 6. c. The relationship between the three genotypes of 302C / T locus and pork quality traits, the model adopted is:

[0052] Y ijkl =μ+G i +S j +N k (+b ijk x ijk )+e ijkl

[0053] Y ijkl is the trait phenotype value, μ is the average value, G i is the genotype effect, S j is the gender effect, N k is the annual effect, b ijk is the regression coefficient of slaughter age, meat quality traits take ...

Abstract

The invention discloses a molecular marker related to pork quality characters and application of the molecular marker, and belongs to the field of molecular markers. The nucleotide of the molecular marker is shown as SEQ ID NO: 1, C / T mutation exists at the 302bp position of the sequence shown as SEQ ID NO: 1, the mutation comprises three mutation types of CC, CT and TT, and when the genotype of a pig individual is CC type, the contents of marbling of longissimus dorsi muscle, marbling of biceps femoris muscle and intramuscular fat of the longissimus dorsi muscle of the pig individual are higher than those of a CT type pig individual and a TT type pig individual. The invention further provides a primer pair for detecting the site, a corresponding detection method is established, and a new molecular marker is provided for genetic improvement of pig muscle quality. According to the present invention, the pork quality character can be subjected to early stage selection, and the detection method is efficient and accurate, such that the pork quality improvement process can be accelerated, and the economic benefits of pig breeding can be improved.

Description

technical field [0001] The invention relates to the field of molecular markers, in particular to a molecular marker related to pork quality traits and its application. Background technique [0002] Pork quality is an important economic trait of pigs. Improving pork quality is one of the important goals of modern pig breeding work, and it is also an important guarantee to meet consumers' needs for pork flavor, food health and nutrition. However, due to the complexity and low heritability of meat quality traits, traditional genetic improvement methods such as phenotype-based selection are inefficient and costly; the emergence of molecular marker-assisted selection (MAS) has greatly accelerated the development of pig breeding. The process, which uses molecular markers associated with specific traits as an auxiliary means for selective breeding, has the advantages of being fast, accurate, and not affected by the environment. It can not only greatly reduce the human and material ...
