Benefits of technology

This patented method allows researchers to find out how well they have been able to detect early stage breast cancer by measuring changes made during or following chemotherapy treatments on their own lung cells. By comparing this data against previous cases, it provides valuable insights into which therapies may be effective at preventing future relapses from developing disease.

Problems solved by technology

The technical problem addressed in this patents relates to developing new therapies or diagnostic markers capable of accurately identifying whether certain types of breast carcinomatosis may develop from other forms of cancer without causing overt symptoms or requiring expensive treatments.

Embodiment 1

[0067] The present invention adopts immunohistochemical method to detect the relative expression of ICOSL protein in breast cancer tissue, and concrete steps are:

[0068] (1) Preparing paraffin sections of breast cancer tissue, and overnight in an oven at 60°C;

[0069] (2) slices are dewaxed to water;

[0070] (Xylene ①10min→Xylene②10min→Xylene③10min→100% ethanol 5min→95% ethanol 5min→85% ethanol 5min→75% ethanol 5min→double distilled water 5min)

[0071] (3) 3% H 2 o 2 Methanol solution, placed at room temperature for 20 minutes;

[0072] (4) Wash with double distilled water for 5min×3;

[0073] (5) Antigen retrieval: Put slices into 0.01M citrate buffer (pH 6.0) and boil for 5 minutes, stop fire for 10 minutes, and then boil for 5 minutes;

[0074] (6) Naturally cool to room temperature, wash with double distilled water for 5min×3;

[0075] (7) Block with 1% BSA for 30 minutes at 37°C;

[0076] (8) Shake off the blocking solution without washing, and directly add th...

Embodiment 2

[0090] Experimental materials: 162 cases of invasive breast cancer surgical specimens and adjacent normal tissue specimens in Shanghai Changhai Hospital from 2003 to 2010

[0091] Method: ICOSL histochemical staining was carried out on the breast cancer tissue specimens and adjacent normal tissues according to the method in the above-mentioned Example 1, and the membrane-expressed ICOSL was scored according to the above-mentioned method.

[0092] Result: if figure 1 As shown in Table 1, the analysis of ICOSL expression in 162 cases of breast cancer and its corresponding adjacent tissues showed that the positive rate of ICOSL in cancer tissues was significantly higher than that in adjacent tissues.

[0093] Table 1: The positive rate and expression level of ICOSL in cancer tissues are up-regulated compared with the corresponding paracancerous tissues

[0094]

Embodiment 3

[0096] Experimental materials: 121 cases of invasive breast cancer surgical specimens and adjacent normal tissue specimens in Changhai Hospital from 2003 to 2010

[0097] Methods: The postoperative treatment was carried out according to the program recommended by the NCCN guidelines, and the survival time was followed up. Considering the expression of ICOSL, age, pathological type, pathological grade, ER, PR, HER2 and other factors, 121 cases of invasive disease were treated. The survival time of breast cancer patients was analyzed by multivariate Cox proportional hazards model.

[0098] Results: As shown in Table 2, considering age, pathological type, pathological grade, ER, PR, HER2 and other factors, the expression of ICOSL in breast cancer cells is an independent prognostic factor for breast cancer.

Abstract

The invention relates to the technical field of biology, in particular to application of ICOSL (Inducible Costimulator Ligand) protein in preparation of a kit used for breast cancer prognosis evaluation. According to the invention, the immunohistochemistry technology and a semi-quantitative analysis method of a staining result are employed to analyze the expression level of the ICOSL protein in atumor tissue, and combine with post-operative follow-up information to determine whether the ICOSL protein expression has correlation with the prognosis of a breast cancer patient or not, wherein theICOSL protein can be used for preparing a protein molecular marker for determining the prognosis of the breast cancer patient to have important guiding significance for post-operative monitoring and sequential treatment of the breast cancer patients.
