[0059] Example 1: In vitro biofilm studies with thermolysin and collagenase
[0060] In vitro assays were performed to demonstrate the bacterial biofilm reducing capacity of thermolysin. In this assay, S. aureus ATCC 6538 was suspended in growth medium of tryptic soy broth supplemented with 0.25% glucose for optimal bacterial biofilm formation. The suspension was transferred to the wells of a sterile 96-well culture plate and incubated at 37°C for 22 hours with one medium change. After the bacterial biofilm was formed, the growth medium was replaced with a thermolysin solution dissolved in the growth medium at different concentrations and a collagenase solution dissolved in the growth medium at the same concentration (enzyme concentration was 0.000001 mg / mL, 0.00001mg / mL, 0.0001mg / mL, 0.001mg / mL, 0.01mg / mL, 0.1mg / mL, 1.0mg / mL and 10mg / mL). After 16 hours, the remaining attached bacteria were quantified by aspirating the enzyme solution (enzyme + growth medium) and washing th...
