[0023] Glycine, disodium hydrogen phosphate, sodium dihydrogen phosphate, Tween-20 and protease inhibitors were all purchased from Sigma-Aldrich, USA. Bovine serum albumin (BSA) was purchased from Merck, Germany. Cy3 fluorescence was purchased from Amerhsma, USA. Other chemical reagents were of analytical grade without further purification before use. All experimental water was ultrapure water treated by Milli-Q50 pure water system (Millipore, USA). Sephadex G-25 column desalting was purchased from GE Healthcare, USA. Chip hybridization kit was purchased from Bio-Rad, USA. Other common glassware are all made in China. Thirty-seven lectins (see Table 1 for specific names) were purchased from Sigma-Aldrich and Vector, Germany, respectively.

Abstract

The invention provides an application of a lectin probe combination to the urine protein glycotype-based gender identification of Qinling rhinopithecus roxellana. The lectin probe combination is a combination of PNA, VVA, BPL, AAL and PTL-I; and if a sample detection result is that PNA and VVA expression is down-regulated and BPL, AAL and PTL-I expression is up-regulated, it is showed that corresponding subjects are the female Qinling rhinopithecus roxellana, otherwise, the corresponding subjects are the male Qinling rhinopithecus roxellana. The lectin probe combination is used for identifyingthe genders of the Qinling rhinopithecus roxellana; and the lectin probe combination is rapid, simple and convenient, accurate and relatively low in implementation cost.
