Problems solved by technology

[0004] The purpose of the present invention is to provide a method for making permanent slide specimens of thrips insects, which can solve the problems of incomplete decolorization of low-concentration lye and cumbersome and time-consuming steps of alcohol dehydration in the production of permanent slide specimens. The specimen has high transparency, long storage time, and the effect of short time required to prepare the specimen

Embodiment 1

[0035] (1) Decolorization: Pick out about 20 thrips insects from the alcohol solution and put them directly into 6% NaOH solution, and place them at room temperature for 8 to 24 hours; the soaking time depends on the size and color of the insects. 1. The soaking time for dark color should be longer; a small number of very light color specimens can be dehydrated directly without decolorization;

[0036] (2) Dehydration: Take the thrips insects out of the lye, put them in distilled water for 2 hours, let the body contents be discharged naturally, and then dehydrate them step by step with alcohol solutions of different concentrations, that is, use 30%, 60%, Soak in 85% and 95% alcohol solution for 5 to 10 minutes respectively, then transfer the specimen to 100% alcohol and leave it for 30 minutes;

[0037] (3) Transparency: Use a straw to suck out the thrips insects and spit them on ordinary filter paper, let the alcohol on the surface of the insects dry naturally, the time shoul...

Abstract

The invention discloses a method for making a permanent slide specimen of thrips. The method comprises the steps: (1) decolorization: taking thrips out of an alcohol solution, putting the thrips intoan alkali solution, and soaking the thrips at room temperature for decolorization; (2) dehydration: taking the decolorized thrips out of the alkali solution, putting the decolorized thrips in distilled water to discharge the contents of the decolorized thrips, and gradually soaking the thrips of which the contents are discharged with alcohol solutions with the concentration progressively increasedfor dehydration; (3) transparency: sucking out the thrips, putting the thrips on filter paper, allowing alcohol on the surface of the thrips to be air-dried naturally until no solvent exists on the filter paper, and soaking the thrips in xylene; and (4) slide sealing: adding dropwise neutral gum or Canadian gum on a slide grass, placing the treated thrips on the neutral gum or Canadian gum, placeall parts of the thrip body, conducting covering with a cover glass, and conducting air-drying to obtain the permanent slide specimen. The obtained specimen has the advantages of high transparency, long storage time and short preparation time.

Description

technical field [0001] The invention relates to a method for making an insect specimen, in particular to a method for making a permanent slide specimen of thrips insects. Background technique [0002] Thrips is a group of insects closely related to human beings, with a wide variety and abundance. Some species of thrips like to inhabit among flowers and have strong pollination ability; many herbivorous species are important crop pests, and a small amount of predatory and fungal-eating species can be used as natural enemy resources. [0003] Thrips individuals are small, and most species are 0.5-5mm in length, so clear and transparent slide specimens are a prerequisite for accurate identification of thrips species. The preparation of thrips permanent slide specimens generally needs to go through steps such as decolorization, dehydration, transparency, and sealing. The control of reagents and time required for each step is an important factor affecting the quality of the speci...
