Abstract

The invention discloses a tissue culture and rapid propagation method of lemon. The tissue culture and rapid propagation method comprises: (1) culture starting: cutting lemon branch with bud stem segment as explant, sterilizing, and inoculating into a starting culture medium, and culturing for 4-5 weeks until axillary buds are sprouted on the explant and grow to achieve a certain length; (2) proliferation culture: cutting the axillary buds obtained in the step (1), inoculating into a proliferation culture medium, and culturing for 4-6 weeks until clustered buds are formed on the base portionsof the axillary buds and grow to achieve a certain length; and (3) rooting culture: cutting the clustered buds obtained in the step (2) into single buds, inoculating the single buds into a rooting culture medium, and culturing for 4-5 weeks until roots are generated and grow to achieve a certain length so as to obtain the lemon tissue culture seedlings. According to the present invention, the culture time of the seedling obtained through the method is short; compared to traditional grafting and cutting, the tissue culture and rapid propagation method of the present invention can significantlyincrease the proliferation coefficient of lemon, increase the survival rate, ensure the genetic stability, and conveniently perform the unified transplanting and planting management at the later period so as to achieve the rapid propagation of lemon.
