Embodiment Construction

[0029] Attached below Figure 1-5 The technical solutions in the embodiments of the present invention are clearly and completely described. Obviously, the described embodiments are only some of the embodiments of the present invention, not all of them. Based on the embodiments of the present invention, all other embodiments obtained by persons of ordinary skill in the art without making creative efforts belong to the protection scope of the present invention.

[0030] The present invention provides the following technical scheme: an experimental method for storing a diluent of sheep semen at room temperature, comprising the following steps:

[0031] 1) Extract the fresh semen of the sheep, and put the extracted fresh semen into the semen collection cup;

[0032] 2) Put the semen-collecting cup filled with semen in step 1) in the water bath of an insulated bucket, and transport the extracted semen back to the laboratory, and prevent the insulated bucket from vibrating violentl...

Abstract

The invention discloses an experimental method for preserving a diluent of sheep semen under a room temperature. The experimental method comprises the following specific steps: 1) extracting the fresh sheep semen; 2) putting a semen collecting cup into a thermal insulation barrel and returning the semen collecting cup to a laboratory; 4) preparing four diluents; 5) putting the diluents into a 37-DEG C water-bath pot; 6) diluting the fresh semen; 7), storing diluted semen I, II, III and IV obtained in the step 6), tightly covering a centrifugal tube cover, putting the centrifugal tube on a foamed centrifugal tube rack, and placing and storing under the room temperature; 8), measuring a sperm motility rate in diluted semen; and 10), detecting a sperm plasma membrane integrity rate in the diluted semen. According to the experimental method for preserving the diluent of the sheep semen under the room temperature, disclosed by the invention, the semen with a relatively high survival rate is extracted, and influences, on experimental results, of a low sperm survival rate are reduced; and survival rates of sheep sperms are separately detected through the four groups of diluents with different proportions, and the measured diluent proportion of the diluent III is more beneficial for survival of the sperms.

Description

technical field [0001] The invention relates to the technical field of an experimental method for storing a diluent of sheep semen at room temperature, in particular to an experimental method for storing a diluent of sheep semen at room temperature. Background technique [0002] Since the invention of the sheep semen preservation technology, it has become one of the most effective ways to improve the utilization rate of high-quality breeding stock and accelerate the improvement of varieties, and it can also reduce the cost of reproduction. The popularization and application of artificial insemination technology has greatly promoted the development of the sheep industry. . The storage methods of semen are divided into cryopreservation, cryopreservation and normal temperature preservation. [0003] Cryopreservation is for the preservation of variety resources and the exchange of germplasm between regions, and can be maintained for a long time; low temperature preservation (0-...
