Problems solved by technology

It has been reported in the literature that different collection methods can affect the identified volatile components
However, for soybean plants at the seedling stage, the effectiveness of these two methods needs to be verified
[0003] At present, there are two defects in the device for collecting soybean plant volatiles: 1) the plant root system is artificially separated from the soil (or substrate), which damages the plant root system to a certain extent, and then affects the release of volatile matter from the plant; The whole set of potted seedlings (together with the plants, plastic pots and their soil or substrate) is placed in a glass container with a source of odor, which brings a lot of impurities, and the volatile components measured according to the device are not actually released by the plants themselves

Embodiment 1

[0028] a, Soybean leaves at the seedling stage sprayed with distilled water

[0029]One day before the collection of soybean volatiles, 10 mL of sterile distilled water was sprayed on the soybean leaves at the seedling stage, and the soybean leaves were neither inoculated nor sprayed with viruses. Soybeans ("Liaoxian No. 1" developed by Liaoning Academy of Agricultural Sciences) were planted in plastic pots (diameter 23 cm, height 18.5 cm) with a mixture of peat and coconut soil (volume ratio of peat and coconut soil: 7:3), 6 plants per pot Plants are used for volatile matter collection when they grow to 8 true leaves (without using any chemical pesticides). The thickness of peat is 20-40mm (developed by PINDSTRUP Company of Denmark), and the thickness of coconut shell is 10-25mm (developed by KLASMANN Company of Germany).

[0035] a. Healthy larvae feed on soybean leaves at the seedling stage sprayed with distilled water

[0036] One day before the collection of soybean volatiles, 10 mL of sterile distilled water was sprayed on the soybean leaves at the seedling stage. After the soybean leaves were dried, 30 mid-3rd instar healthy larvae (starved for 12 h) were picked up on the soybean leaves, and let them grow in the soybean leaves. The leaves were used for volatile matter collection after feeding on the leaves for 24 hours. [0042] a. Susceptible larvae feed on soybean leaves at the seedling stage sprayed with distilled water

[0043] One day before the collection of soybean volatiles, 10 mL of sterile distilled water was sprayed on the soybean leaves at the seedling stage. After the soybean leaves were dried, 30 infected third-instar mid-stage larvae (starved for 12 h) were placed on the soybean leaves. It was used for volatile matter collection after feeding on soybean leaves for 24 hours. Soybeans ("Liaoxian No. 1" developed by the Liaoning Academy of Agricultural Sciences) were planted in plastic pots (diameter 23 cm, height 18.5 cm) of a mixture of peat and coconut soil (volume ratio of peat and coconut soil 7:3), 6 plants per pot Plants are used for volatile matter collection when they grow to 8 true leaves (without using any chemical pesticides). Abstract

The invention discloses a method and a device for collecting infectious pest feeding induced seedling-stage soybean plant volatile matters. The method comprises the following steps: wrapping a plastic pot of a seedling-stage soybean plant sample to be tested with aluminum-foil paper so as to eliminate influence of a mixture of coconut soil and turf and volatile matters released from the root of a soybean plant; covering plants to be tested by using a cylindrical glass cover, and covering the upper opening of the glass cover by using sterile clean stainless steel piece; inserting an extraction head of a solid-phase micro extractor into the glass cover through a round hole in the stainless steel piece, continuously enriching the volatile matters for 30 minutes, and taking out the extraction head; performing thermo-analysis for 2 minutes at 230 DEG C, and performing gas chromatography-mass spectra (GC / MS) combined analysis. The device comprises the planting pot, the cylindrical glass cover, a stainless steel thin ring, the aluminum-foil paper, the solid-phase micro extractor, the plant and a base stock medium for cultivating the plant. By adopting the method and the device, the seedling-stage soybean plant volatile matters can be accurately detected, and the volatile matters which are accurately released from the plant can be accurately tested.

Description

technical field [0001] The invention belongs to the field of biological control of pests, and in particular relates to a method and a device for collecting volatiles of soybean plants at the seedling stage induced by poison-susceptible insects. Background technique [0002] Compared with soybean plants at flowering, pod-setting and bulging stages, plants at the seedling stage released less volatile matter and were difficult to collect. Collecting volatiles is an essential stage in the early stage of identifying volatile components. For this reason, designing a reasonable volatile collection method and collection device is particularly important for the scientific identification of volatile components in soybean plants. Solid-phase microextraction and solvent extraction are two typical methods for collecting volatiles from herbal plants. It has been reported in the literature that different collection methods can affect the identified volatile components. However, for soyb...
