Embodiment 1

[0037] (1) On the premise of signing the informed consent form with the patient, all samples were taken from fasting venous blood anticoagulated with heparin in the morning. Peripheral blood samples from 30 patients with newly diagnosed untreated AML and 12 healthy people were collected. This part of the research protocol has been approved by the unit ethics committee. At the same time, clinical data such as clinical curative effect of AML patients were collected (as shown in Table 2).

[0038] (2) Separation of peripheral blood mononuclear cells. Take 4ml of lymphocyte separation medium (Ficoll, density 1.077) and add it into a 15ml centrifuge tube, spread the diluted anticoagulated peripheral blood sample suspension on the separation medium, and centrifuge at 1500rpm for 15 minutes in a horizontal centrifuge. Transfer the single nuclear layer in the middle to another 15ml centrifuge tube, then add an appropriate amount of 1×PBS, blow gently, centrifuge and wash at 1000rpm ...

Abstract

The invention discloses an application of a TCR[gamma][delta]T cell subfamilies in preparing a kit for predicting a curative effect and prognosis evaluation of AML (acute myeloid leukemia). The invention is based on the technical scheme which is proposed when the inventor of the invention discovers a relationship between the clonal proliferation situation of peripheral blood [gamma][delta]T cell TRDV1-RTDV8 subfamilies of a patient with the AML and the curative effect and the prognosis of the patient with the AML for the first time. The clonally-proliferated TRDV4 and TRDV8 subfamilies, as protective factors, are related to the complete remission of the patient with the AML, and the clonally-proliferated TRDV5 and the TRDV6 subfamilies are related to the recurrence of the patient with the AML. The pedigree analysis and clonal proliferation situation of the TCR[gamma][delta]T cell subfamilies have a broad prospect in the aspects of predicting the clinical curative effect and prognosis evaluation of the patient with the AML, and more fundamental research data can be provided for the individualized treatment of the AML.

Description

technical field [0001] The invention belongs to the field of biomedicine, and particularly relates to the application of the TCRγδT cell subfamily in the preparation of a kit for predicting the curative effect and prognosis of AML. Background technique [0002] Acute myeloid leukemia (AML) is a group of malignant clonal diseases originating from hematopoietic stem cells. Leukemic cells stagnate at different stages of cell development due to the loss of the ability to differentiate and mature, and there are a large number of them in bone marrow and other hematopoietic tissues. Accumulation inhibits normal hematopoiesis. Recent studies have shown that γδT cells have good application prospects in the adoptive immunotherapy of AML, but there are no laboratory indicators related to γδT cells that can effectively predict the clinical efficacy and prognosis of AML patients. Contents of the invention [0003] The purpose of the present invention is to overcome the shortcomings an...
