Abstract

The invention discloses a tissue culture seedling medium rooting method which comprises the following steps: (1) by using a tissue culture seedling propagation technique, separating cluster buds propagated from a solid propagating medium, of American red maple, and culturing in a seedling strengthening culture medium, so as to obtain rootless tissue culture seedlings of the American red maple; (2) soaking jiffy seedling growing blocks in a liquid rooting culture medium, taking out the jiffy seedling growing blocks, leaving to stand, draining off the water, putting into culture bottles, sealing, and sterilizing; (3) cutting off bottom stems of the tissue culture seedlings of the American red maple obtained in the step (1), and inserting into the seedling growing blocks for culture in the step (2) so as to obtain rooted sterile tissue culture seedlings; (4) acclimatizating and transplanting the sterile tissue culture seedlings in a greenhouse so as to obtain sterile tissue culture seed seedlings; (5) transferring the sterile tissue culture seed seedlings into hole trays for culture, thereby obtaining seedlings. As the JIFFY seedling growing blocks are adopted for rooting induction on the sterile tissue culture seedlings in the method, the labor cost can be lowered, the seedling consumption and the acclimatization difficulty can be reduced, a step of seedling transplantation can be canceled, and the seedlings are high in survival rate and can grow strongly.
