[0033]Example Ⅱ, Arthrobacter sp. ZS3 Degradation DDT and HHO Quantitative Determination

[0034] 1. Preparation of standard curves for allelochemicals DDT and HHO

[0035] DDT and HHO standard solutions with concentrations of 5, 20, 80, 120, and 200 μg / mL were prepared with HPLC grade methanol. Each concentration was injected 3 times, the results were averaged, the peak area was plotted against the concentration, and the standard curve was drawn.

[0036] 2. Determination of the ability of ZS3 strain to degrade DDT and HHO

[0037] The strain ZS3 was inserted into 40mL nutrient medium for expansion for 2 days, centrifuged at 4000r / min for 5 minutes, the precipitate was suspended in 1mL sterile water, and the concentration of the bacterial suspension was determined by plate counting method. Take 100uL of the bacterial solution and insert it into 10mL of inorganic salt culture solution containing 200ug of DDT and HHO, culture on a shaking table for 5 days, and take samples to...

Abstract

The invention discloses a biocontrol bacterial strain for reducing allelopathy of Eupatorium adenophorum and application thereof. The biocontrol strain is Arthrobacter sp. ZS3, which is preserved in the General Microorganism Center of China Committee for the Collection of Microorganisms, with the preservation number CGMCC NO.9283. The Arthrobacter ZS3 of the present invention can degrade 49.8% and 56.8% of euplendione and hydroxyzelenone with a concentration of 100ug / mL in inorganic salt medium within 5 days. The allelopathic effect of Eupatorium adenophorum was reduced by 46.8% after spraying ZS3 bacterial agent with a concentration of 109-1010 CFU / mL at the ratio of 4mL / 100g soil. This shows that the strain can effectively degrade allelochemicals and has broad application prospects in the control of Eupatorium adenophorum.

Description

technical field [0001] The invention belongs to the technical field of plant protection, and in particular relates to a biocontrol bacterial strain capable of effectively reducing the allelopathic effect of the alien invasion plant Eupatorium adenophorum. At the same time, the present invention also relates to a method for using the biocontrol bacterial strain in biological control. Background technique [0002] Ageratina adenophorum (Ageratina adenophorum) is a vicious perennial invasive alien weed of Compositae. It is native to Mexico and Costa Rica in Central America, and is now widely distributed in more than 30 tropical and subtropical countries and regions in the world. The invasion of Eupatorium adenophorum has caused serious impact on the local ecological environment, mainly in crowding out the growth of other local plants, forming a single superior population, and destroying biodiversity. After Eupatorium adenophorum invades farmland, forest land, and pasture, it c...
