Abstract
The invention discloses a nucleic acid aptamer specifically combined with β-amyloid precursor protein lyase 1 and application thereof. The nucleotide sequence of the nucleic acid adapter is any one shown in SEQ ID NO:1 or SEQ ID NO:2. The use of the nucleic acid aptamer or its chemical modification as a BACE1 inhibitor in the preparation of drugs for treating Alzheimer's disease. In this study, DNA aptamers A1 and A4 that can bind to BACE1 with high affinity were successfully screened by SELEX technology, and it was proved that the aptamers could inhibit the activity of BACE1 in vitro. It provides a research basis for the development of new potent and specific BACE1 inhibitors, and provides new methods and ideas for AD treatment.
Description
[0001] Technical field: [0002] The invention belongs to the field of biomedicine, and specifically relates to a nucleic acid aptamer specifically combined with β-amyloid precursor protein lyase 1 and an application thereof. [0003] Background technique: [0004] Alzheimer's disease (AD) is a progressive degenerative disease of the central nervous system, with cognitive dysfunction as the main clinical manifestation, and other symptoms including executive dysfunction and mental disorders. Advanced patients cannot take care of themselves and often die from complications such as lung infection; it brings huge economic burden to the patient's family and society [1] . It is predicted that the number of AD patients worldwide will triple from 36 million in 2010 to 2050 [2] . The cause of AD is unknown. At present, it is believed that a variety of pathogenic factors, such as amyloid precursor protein / amyloidβ (APP / Aβ) [3] , Apolipoprotein E4 (ApolipoproteinE4, apoE4) [4,5] 、ta...
