Problems solved by technology

[0003] In repeated experiments conducted in our laboratory, adventitious buds could not be induced from leaves of tested bird's nest fern species, indicating that this technique is only applicable to some species, so it is necessary to develop tissue culture techniques for different plant regeneration pathways of bird's nest fern

[0029] 1. Materials and Methods

[0030] 1.1 Test material

[0031] Bird's nest fern (Asplenium nidus) belongs to Aspleniaceae (Aspleniaceae) fern, and its seedlings were purchased from Nanjing Huambo Flower Market. The laboratory collects mature spores from the leaves of potted plants, and obtains sterile seedlings by using tissue culture methods (Qin Tinghao, Zou Zonglan. Tissue Culture of Bird's Nest Fern [J]. Plant Physiology Communications, 2004,40(3):349).

[0033] MS improved medium consists of MS macroelements, MS trace elements (MurashigeT, SkoogF.A revised medium from rapid growth and bioassays with tobacco tissue cultures[J].Physiol plant,1962,15:473~497) and B 5 Vitamin (Gamborg O L, Miller R A, Ojima K. Nutrient requirements of suspension cultures of soybean root cells [J]. Exp. Cell Res., 1968.50:151~158), a commonly used medium in plant tissue culture, sucrose 30g / L, activated carbon mass ratio 0.1% (Nanjing Zhuyan Biotechnology Co., Ltd.)...

Abstract

The invention discloses a regeneration method for small green ball plants by using induction of roundleaf new pteris fern and belongs to the technical field of plant cultivation methods. Apical short stems of roundleaf new pteris fern germfree seedlings are used as explant materials; small green balls are induced and generated on an MS improved culture medium added with phenyl urea cytokinin CPPU of 1.5-2.5mg / L and indolebutyric acid IBA of 0.1mg / L; the induction frequency of the small green balls is up to 100 percent. Multiplication of the small green balls is implemented on the MS improved culture medium added with the CPPU of 1.0-1.5mg / L and the IBA of 0.1mg / L; the multiplication coefficient of the small green balls of each generation is 5. The small green balls are germinated on the MS improved culture medium added with the CPPU of 0.2-0.5mg / L and the IBA of 0.1mg / L to form seedlings. The seedlings grow into small complete plants with roots, stems and leaves on the MS improved culture medium added with the IBA of 0.2-0.4mg / L, and the adult rate is over 95 percent. The method is suitable for new pteris fern test tube seedling industrial production.

Description

technical field [0001] The invention relates to a high-frequency plant regeneration method induced by bird's nest fern to induce green pellets, and belongs to the technical field of plant cultivation methods. Background technique [0002] Up to now, among the 12,000 species of ferns, only a small part have been successfully tissue cultured. Although some genera and species have been successfully tissue cultured, they have the problems of long reproductive cycle and low proliferation rate (Han Jing, Zhao Li. Fern Advances in research on plant-like reproduction [J]. Anhui Agricultural Sciences, 2005, 33(7): 1261-1263). Domestic bird's nest fern tissue culture technology research is to propagate test-tube seedlings through the differentiation of seedlings and proliferation of seedling clusters, induction of adventitious buds and proliferation. Using young leaves of bird's nest fern as explant material, the induction rate of adventitious buds reached 85%, and the proliferation ...
