According to the invention, urine samples of female giant pandas at a spring breeding period are collected and are subjected to chemical analysis; conditions such as a time for an estradiol maximal peak or a subsequent estradiol peak to decrease with an amplitude reaching a value above 50%, whether a progesterone value is in an increasing trend during the decreasing period of the estradiol maximal peak or the subsequent estradiol peak, whether the giant pandas show a typical estrous peak behavior, and a ratio of the corresponding progesterone value of the estradiol maximal peak or the subsequent estradiol peak to a progesterone basic value are combined, and the giant panda estradiol true peak value is determined. The method provided by the invention is obtained on the basis of statistical analysis of estradiol variation regularities of 78 oestrus giant pandas which constructs a maximal datum quantity currently, such that the method has high reliability.
