AI Technical Summary

Benefits of technology

This technology allows for better detection of New Buganese (BV) viruses by measuring their ability to produce light when exposed to certain substances or conditions that can affect how they infect other animals. It also shows promise at very low concentrations compared with existing methods such as ELISA but this effectiveness depends on factors like temperature and humidity levels.

Problems solved by technology

The technical problem addressed in this patented text relates to finding ways to detect novel feline coronas (focal leukocyte common cold) causing illnesses like dengue hemorrhagic fevers and chikungunya attacks during springtime periods when people live without exposure to these harmful agents. Current diagnostic tools require long hours before they become sick enough to respond quickly but cannot accurately identify any potential sources of contamination until after their occurrence. Therefore, researchers aimed towards developing faster and more efficient techniques to help fight against future threatening events related to New Bantany Coronaria Virus (NBV).

Embodiment 1

[0039] 1 Materials and methods

[0040] Virus strains and clinical specimens:

[0041] The new bunyavirus strain is an isolate from the Zhejiang Provincial Center for Disease Control and Prevention, and its DNA is shown by the fourth sequence in the sequence list. The clinical samples come from the serum of suspected patients with a new type of bunyavirus outbreak in Zhejiang Province recently. After the samples are collected, they are transported to the laboratory on ice.

[0042] 1.2 Primers and probes

[0043] Genome sequences of novel bunyavirus strains from across the country were downloaded from the NCBI GenBank in the United States. Homology comparisons were carried out, and new-type bunyavirus-specific primers and Taqman probes were designed in the L gene region of all novel bunyavirus genomes. The sequences are as follows (sequences 1 to 3 in the sequence table):

[0044] Upstream primer: 5'-AGGTTTGAGATATTCCCCAAGACA-3'

[0045] Downstream primer: 5'-AATTTTTGTAGAAG...

Abstract

The invention relates to a novel orthobunyavirus fluorescence quantitative RT-PCR (Reverse Transcriptase-Polymerase Chain Reaction) detection kit and a novel orthobunyavirus detection method, and aims to provide the kit which is convenient to use and accurate in detection and the detection method which is high in accuracy and high in detection efficiency. The technical scheme is that: the novel orthobunyavirus fluorescence quantitative RT-PCR detection kit comprises a specific primer, a fluorescent probe, a PCR buffer solution, dideoxyribonucleotide triphosphate and a mixed preparation, wherein an upstream primer has a sequence of FP:5'-TGGCCCTGGCTCTATAAACAT-3'; a downstream primer has a sequence of RP:5'-AATGGCAGCCCAAATGAATC-3'; the specific probe has a sequence of P:5'-FAM-TCCAATGAYGCCAAGAAGTGGAA-BHQ1-3'; FAM is a fluorescent reporter group; and BHQ1 is a fluorescent quenching group.
