Embodiment 1

[0021] 1. Extract the plasmid containing the sucrose-6 fructose transferase (sacB) gene

[0022] Purchase a strain of Escherichia coli (E.coli) DH5F' (ATCC#77281), which contains the plasmid pHSS21, which has a hygromycin resistance marker and a sucrose-6 fructose transferase (sacB) gene, and thus is sensitive to sucrose sex. See (Schweizer, H.P. (1992) Pseudomonas cyanides allelic exchange using Coli1-type vectors and cassettes with portable oriT and sacB markers. Molecular Microbiology 6(9) 1195-1204). Using conventional molecular biology methods, the plasmid pHSS21 was extracted and used as the plasmid used in the next step of transformation.

Abstract

In the preparation process of cow mastitis vaccine, Gram-negative bacterium containing core universal antigen is utilized and transformed with genetic plasmid containing sucrose-6 fructose transferase. Under the induction of sucrose, Gram-negative bacterium suicides and the Gram-negative bacterium with core universal antigen is collected to prepare the cow mastitis vaccine. The said process produces no damage to immunogenicity of the vaccine and no substituted or non-natural vaccine antigen. Using non-toxic sucrose as chemical inducing matter can reduce the endotoxin in and side effect of the vaccine. The vaccine stimulates the immunological system to resist coibacillus, salmonella and other Gram-negative bacteria causing cow mastitis.

Description

technical field [0001] The invention relates to a preparation method of a vaccine, in particular to a preparation method of a bovine mastitis vaccine. Background technique [0002] Bovine mastitis is a serious and costly disease on dairy farms around the world. This disease usually has the possibility of onset at various stages of lactation, and 50% of bovine mastitis occurs in the early stage of lactation. At all stages of lactation, mammary gland secretions are a good medium for bacterial growth. When external bacteria successfully pass through the nipple and ducts and enter the mammary gland tissue, it will cause infection and inflammation in the mammary gland. [0003] Cases of clinical bovine mastitis are caused by bacteria in the environment, and the overall percentage of bacterial mastitis incidence has risen sharply in recent years. Bacteria are microorganisms that exist in the environment. However, after the cow gives milk, since the pores of the mammary gland ar...
