Benefits of technology

"The present invention provides delivery constructs for administering antibodies to a subject. These delivery constructs include a carrier construct that non-covalently binds to the antibody or antibody fragment of interest. The carrier construct can be a fusion protein comprising a receptor-binding domain, a transcytosis domain, and an antibody-binding domain. The carrier construct can also have multiple antibody-binding domains separated by a cleavable linker. The antibody-binding domain can be specific to the antibody or antibody fragment of interest. The delivery constructs can be used to treat various conditions in a subject."

Problems solved by technology

However, administration of these therapeutic antibodies remains problematic.
Such injections require penetration of the subject's skin and tissues and are associated with pain.
Further, penetration of the skin breaches one effective nonspecific mechanism of protection against infection, and thus can lead to potentially serious infection.

5.1. Definitions

[0025]Unless defined otherwise, all technical and scientific terms used herein have the meaning commonly understood by a person skilled in the art to which this invention belongs. As used herein, the following terms have the meanings ascribed to them unless specified otherwise.

[0026]A “ligand” is a compound that specifically binds to a target molecule. Exemplary ligands include, but are not limited to, an antibody, a cytokine, a substrate, a signaling molecule, and the like.

[0027]A “receptor” is compound that specifically binds to a ligand.

[0028]“Immunoassay” refers to a method of detecting an analyte in a sample involving contacting the sample with an antibody that specifically binds to the analyte and detecting binding between the antibody and the analyte. A variety of immunoassay formats may be used to select antibodies specifically immunoreactive with a particular protein. For example, solid-phase ELISA immunoassays are routinely used to select monoclonal antibod...

Abstract

The present invention relates, in part, to methods and compositions for needleless delivery of antibodies to a subject. The present invention also relates, in part, to methods for needleless delivery of fusion proteins comprising a bioactive molecule and an antibody fragment to subject. In one aspect, the methods and compositions involve administering to the subject a delivery construct comprising a carrier construct non-covalently bound to the antibody or fusion protein to be delivered, wherein the carrier construct comprises a receptor-binding domain, a transcytosis domain, and an antibody-binding domain to which the antibody or the antibody fragment of the fusion protein non-covalently binds.

Description

1. FIELD OF THE INVENTION[0001]The present invention relates, in part, to methods and compositions for needleless delivery of antibodies to a subject. The present invention also relates, in part, to methods for needleless delivery of fusion proteins to subject, the fusion proteins comprising a bioactive molecule and an antibody fragment. In one aspect, the methods and compositions involve administering to the subject a delivery construct comprising a carrier construct non-covalently bound to the antibody or fusion protein to be delivered, wherein the carrier construct comprises a receptor-binding domain, a transcytosis domain, and an antibody-binding domain to which the antibody or the antibody fragment of the fusion protein non-covalently binds.2. BACKGROUND[0002]Advances in biochemistry and molecular biology have resulted identification and characterization of many therapeutic antibodies. Administration of antibodies can result in drastic improvements in quality of life for subjec...
