Abstract

The invention discloses a fluorescence ternary correlation spectrum system for analysis of binding interaction among three molecules. The fluorescence ternary correlation spectrum system comprises at least three excitation light sources outputting different wavelengths, the excitation light sources illuminate fluorescence signals generated by excitation of a sample, and the fluorescence signals are divided into multiple paths of signals according to fluorescence wavelengths. A signal is transmitted to the signal acquisition card and the operation card through detection of the single-photon detector, and ternary correlation function real-time operation is carried out to obtain a fluorescence ternary correlation spectrum curve. According to the system, a fluorescence ternary correlation spectrum curve is obtained by performing real-time recording and ternary correlation function analysis on fluorescence fluctuation signals of three kinds of fluorescence molecules emitting different wavelengths in a detection micro-area through a fluorescence ternary correlation spectrum, and the relationship between the amplitude of the curve and the concentration of three kinds of fluorescence molecule compounds is analyzed; the analysis of the joint binding effect among three different molecules is realized, and the in-situ analysis research of a fluorescence correlation spectrum in living cells is expanded.

Description

technical field [0001] The invention relates to a fluorescence ternary correlation spectrum system used for analyzing the combination of three molecules, belonging to the field of spectrum analysis. Background technique [0002] Microscopic imaging and spectral analysis technologies have become important research tools in current life science research due to their advantages of non-invasiveness and high detection sensitivity, such as photoactivated localization microscopy and stochastic optical reconstruction microscopy. Through continuous pairwise and ternary interactions between different biomolecules in cells, electron transfer, signal transduction, function regulation, etc. are carried out to realize the physiological functions of cells. The occurrence of abnormal intermolecular interactions in cells, such as the aggregation of polyproteins, is the basis of many diseases, such as Creutzfeldt-Jakob and Alzheimer's disease, and may lead to cancer. Many technologies curren...
