Problems solved by technology

After decades of development, surgery, radiotherapy and chemotherapy have not significantly improved the 5-year survival rate of head and neck cancer patients, and due to the special anatomical site, the resection range of head and neck cancer surgical treatment is severely limited, and the quality of life of patients after treatment generally declines , often disfiguring or disabling

Embodiment Construction

[0015] The present invention will be further elaborated below through specific examples. It should be noted that the following descriptions are only for explaining the present invention, and do not limit its content.

[0016] The method for obtaining the molecular marker of the present invention is as follows:

[0017] 1. Download 620 tRNA gene annotation information and their sequences from the GtRNAdb (http: / / gtrnadb.ucsc.edu / genomes / eukaryota / Hsapi19) database. Annotations and sequence information for newly constituted CCA-tRNAs were obtained by adding "CCA" at the 3′ end of the mature tRNA sequence. Then, starting from the 5' or 3' end of the CCA-tRNAs sequence, all fragment sequences in the range of 15-30 nt were obtained. After further merging the same sequences, 8681 fragment sequences and 9285 fragment sequences from the 5' end of CCA-tRNAs were finally obtained. A fragment sequence from the 3′ end of CCA-tRNAs. In addition, we used the bedtools tool to obtain the an...

Abstract

The invention discloses a novel molecular marker and its application in preparation of a kit for the diagnosis and prognosis of head and neck cancer. The RNA sequences of the novel molecular marker are shown in SEQ ID NO:1-SEQ ID NO:48. Based on the marker of the invention, a mathematical model for the diagnosis of head and neck cancer is constructed; the model has high sensitivity, good specificity, AUC up to 0.981, and good diagnostic effect. In addition, these 48 tRF fragments can also be used as molecular markers for head and neck cancer classification and prediction of patient survival; in the test data, according to the expression of tRFs, head and neck cancer tumor samples were clustered into 4 subtypes, and analyzed by survival curves It shows that there are significant differences in the survival periods of these subtypes; the novel tRF molecular marker disclosed in the present invention has the characteristics of good diagnostic indicators, can be effectively used in the diagnosis, classification and prognosis of head and neck cancer, and has high clinical application and promotion value.

Description

technical field [0001] The invention relates to the technical field of biological detection, in particular to a novel molecular marker and its application in preparing a kit for diagnosis and prognosis of head and neck cancer. Background technique [0002] Head and neck cancer includes tumors originating from any tissue or organ in the head and neck except the eye, brain, ear, thyroid and esophagus, including neck tumors, ENT tumors, and oral and maxillofacial tumors (oral cancer). Since more than 90% of head and neck tumors are squamous cell carcinomas, head and neck cancers are also called head and neck squamous cell carcinomas (HNSC). Head and neck cancer is a malignant tumor with a high incidence in China and even the global cancer spectrum. It ranks sixth among common tumors. It has a high degree of malignancy and an average 5-year survival rate of less than 50%. After decades of development, surgery, radiotherapy and chemotherapy have not significantly improved the 5-...
