Problems solved by technology

At present, composite pH microelectrodes are commonly used to measure the pH value of plant body fluids in situ, but it is difficult to achieve in situ determination for some relatively thin or small organ samples such as leaves and stems, and some woody and hard plants are easy to cause the test The instrument is broken (such as the tip of the pH microelectrode), so it is imminent to establish a simple method that can realize the pH value of plant body fluid outside the device, which can make the pH value of plant organ body fluid as an effective indicator method or surrogate parameter , for applied research in the fields of plant physiology, plant nutrition or biogeochemistry

Embodiment 1

[0025] (1) The air-dried or dried plant organ samples were repeatedly washed with tap water and deionized water for 5 times each, dried in a blast drying oven at 40°C, then pulverized with a plant grinder, and passed through a 60-mesh analytical sieve.

[0026] (2) Investigation of the volume of the soaking solution: Weigh 0.5 g of the plant sample powder in step (1) accurately, place it in a polytetrafluoroethylene crucible, and add 2, 3, 4, and 5 mL of deionized water to it, so that the deionized The sample powder was completely submerged in ionic water, left to soak for 30 minutes, and the soaked leachate was filtered out, and the compound pH microelectrode was used to measure the leachate, and the change curve of pH value was as follows: figure 1 shown. The results showed that the volume of deionized water had no significant effect on the determination of pH.

Abstract

The invention discloses an ex-situ determination method for the pH value of plant sap. The ex-situ determination method comprises the following steps: washing an air-dried or kiln-dried plant organ sample with tap water and deionized water five times respectively, then drying the plant organ sample in a drying box at 40 DEG C, then crushing the plant organ sample with pulverizer and carrying out screening with a 60-mesh sieve; accurately weighing 0.4 to 0.6 g of powder obtained in the previous step, placing the powder in a teflon crucible, then adding 2 to 5 mL of deionized water to allow the powder to be totally immersed in the deionized water and then carrying out standing and immersion for 30 to 60 min; and filtering out leachate and determining the pH value of the leachate by using a pH electrode so as to obtain the pH value of plant sap. The method provided by the invention overcomes the technical problem that the pH value of plant sap can only be determined in situ in an organ and provides technical guarantee for application of the pH value to research of fields like phytophysiology, plant nutriology and biogeochemistry.

Description

technical field [0001] The invention relates to a method for measuring the pH values of body fluids of different organs outside the device. Background technique [0002] In recent years, with the improvement and innovation of analytical technology, the methods for measuring pH value have been continuously introduced, including acid-base indicator method based on agar or glass powder, weak acid flux method, composite pH electrode method, fluorescence spectroscopy, confocal Laser scanning microscopy, microelectrode pH sensors and other methods. The establishment of these methods provides a good technical basis for the determination of the pH value of plant body fluid. At present, composite pH microelectrodes are commonly used to measure the pH value of plant body fluids in situ, but it is difficult to achieve in situ determination for some relatively thin or small organ samples such as leaves and stems, and some woody and hard plants are easy to cause the test The instrume...
