The method comprises the following steps: collecting a specimen, and collecting 2-5ml of sputum of a subject; preparing a treating liquid, and preparing a sputum pretreating liquid for inspecting acid-fast bacilli by the liquid-based thin-layer cell smears; centrifuging, adding the sputum into the prepared sputum pretreating liquid, and vibrating for multiple seconds; preparing a section, absorbing multiple milliliters of the sputum at the bottom of a sputum pretreating liquid test tube by a sucker, putting the sputum into a section preparation bin, putting the section preparation bin into a section preparation machine, and taking out the section preparation bin to dry when the section preparation machine completes the preparation; and carrying out acid-fast staining and microscopic inspection.
