Abstract

The invention discloses a method for obtaining explants for tissue culture of bamboo. The method comprises the steps: selecting tested fine branch stumps of bamboo groves, and planting the branch stumps in a container; cutting off the mother bamboos growing up in current year at a distance of 1.3-1.8m to the ground in August to October; applying fertilizer and trimming regularly and cutting the roots according to conditions so as to control the growth of the bamboos; clipping half-lignified branches from the mother bamboos for a plurality of times in May to November annually, cutting off the leaves and trimming the branches into stems; washing and disinfecting the stems through liquid detergent, tap water, alcohol, corrosive sublimate and sterile water; and finally trimming the stems again to obtain sterile explants for tissue culture of bamboo. Through the method, explants in large quantity with good quality and low contamination rate can be obtained, and the cluster buds have strong germination ability. The method is simple and convenient in operation and low in production cost. The method is suitable for industrialized seedling production of bambusa texlitis, bambusa chungii and dendrocalamus minor, and has good economic, social and ecological effects.
