[0020] DDR1 is a member of the receptor tyrosine kinase family, and its ligand is collagen. DDR1 plays an important role in tissue fibrosis and is an attractive anti-fibrotic target. Studies have shown that collagen activates DDR1 to aggregate and enhance its binding to collagen, and DDR1 allows collagen remodeling to promote fibrosis. After collagen binding, DDR1 can be shed through the mediation of membrane-anchored membrane-type collagenase (MT) 1, MT2 and MT3 matrix metalloproteinase (MMP). Collagen causes specific stimulation of the shedding of the extracellular end of DDR1, but does not affect other transmembrane proteins. Collagen-stimulated shedding of the extracellular DDR1 domain has been demonstrated in many studies. Collagen is the most abundant extracellular matrix in the human body. In tissue fibrosis, increased deposition, alignment, and crosslinking of localized collagen fibers lead to tissue hardening. In the dynamic synthesis of extracellular matrix, incl...
