Abstract

The embodiment of the present invention relates to modified T cells comprising antigen binding molecules, wherein the expression and/or functions of CDC42 in the modified cells is enhanced. In embodiments, the modified cell antigen binding molecules have increased cytokine release level in response to the antigen to which they bind as compared to corresponding T cells that do not overexpress the CDC42. In embodiments, cytokine release includes the release of IFN gamma cytokines. In embodiments, the modified cells have enhanced migration ability to chemokines as compared to corresponding T cells that do not overexpress the CDC42.

Description

technical field [0001] The present disclosure relates to compositions and methods related to chimeric antigen receptor cells and their use in the treatment of diseases including cancer. Background technique [0002] Cancer, known as a malignant tumor, involves abnormal cell growth and has the potential to invade or spread to other parts of the body. In humans, there are more than one hundred types of cancer. Often, once cancer cells break off, they spread throughout the body through the blood and / or lymphatic system, becoming life-threatening. Currently, CAR-T therapy does not appear to be effective against solid cancers. One of the challenges is that CAR-T cells don't seem to be able to efficiently migrate to these cancer cells that have spread into the body. Contents of the invention [0003] Embodiments relate to T cells comprising a modification of an antigen binding molecule, wherein the expression and / or function of CDC42 in the modified cell has been enhanced.
