Problems solved by technology

The success of RNA interference mainly depends on the selection of target genes. At present, the research on insects mainly focuses on the enzymes related to nerve, metabolism and reproductive development, and there are few studies on the synthesis of juvenile hormone.

Embodiment 1

[0021] Embodiment 1, prevent and treat Manduca sexta

[0022] (1) Insect sources and feeding methods,

[0023] The eggs of the tobacco hornworm (Manduca sexta) were purchased from Carolina Biological Supply Company (Burlington, NC), and were artificially raised in a light incubator with a photoperiod of L16:D8, a rearing temperature of 26±1°C, and a relative humidity of 70%. ~90%. The larvae were fed with a special feed for Manduca sexta, and the adult moths were fed with a sucrose liquid feed (sterilized at 121° C. for 20 min, 8% sucrose aqueous solution). When the larvae reach the ecdysis line at the end of the fifth instar, they are transferred to the light-shielding small tube alone. The larvae stop taking food and molt into pupae in the next day. The pupae are transferred to the eclosion box, which has long-term light and places tobacco or other Solanaceae plants, and soon the pupae emerges as adults for mating and laying eggs.

Embodiment 2

[0052] Embodiment 2, control cotton bollworm

[0053] (1) Insect sources and feeding methods,

[0054] The cotton bollworms used in the present invention are purchased from Keyun Biology, and then bred and propagated in the laboratory.

[0055] The feed is artificial feed, which is configured by our laboratory. The water source is tap water. The feeding conditions are 28±1°C, the relative humidity is 50%-70%, and the photoperiod is L14:D10.

Abstract

The invention discloses a method for effective prevention and control of lepidoptera pests by RNAi. By means of feeding, double-stranded RNA (namely dsRNA) aiming at farnesol dehydrogenase FOLD, juvenile hormone transmethylase JHAMT and juvenile hormone epoxidase CYP15C1 related genes is introduced into larva bodies to realize prevention and control of the lepidoptera pests. The method is used forprevention and control of the lepidoptera pests, and high specificity in prevention and control, durable and safe prevention and control effects and freeness of harms to mammals and the like are realized.

Description

technical field [0001] The invention relates to the technical field of biological control, in particular to a method for effectively controlling lepidopteran pests by using RNAi. Background technique [0002] Insects of the order Lepidoptera mainly include moths and butterflies. About 200,000 species are known in the world, and more than 8,000 species are known in China. Among them, there are 6,000 species of moths and 2,000 species of butterflies. It is also an order with the most agricultural and forestry pests. Most types of larvae damage all kinds of cultivated plants, and the larger ones often eat up the leaves of plants or drill into branches. Smaller ones tend to curl leaves, adorn leaves, form sheaths, spin silk and form nets, or drill into plant tissues to feed and cause damage. The vast majority of Lepidoptera larvae are terrestrial, mostly herbivorous, and are widely distributed worldwide. At present, physical trapping and chemical insecticides are mainly use...
