Problems solved by technology

Through the observation of TAC1 overexpression and RNAi transgenic lines, it was found that the up-regulation of TAC1 would lead to an increase in the tiller angle of rice plants, while the down-regulation of TAC1 would lead to a decrease in the tiller angle.

[0040] 1. Cloning of rice OsRUS1 gene

[0041] Primers were designed according to the cDNA sequence of the OsRUS1 gene provided by the MSU / TIGR rice genome database (http: / / rice.plantbiology.msu.edu / ), OsRUS1 ORF-F1: (5'-A GGATCC ATGTCCTCCTCGCAATCTCT-3′, the underline is the restriction site of BamHI) OsRUS1 ORF-R1 (5′-G AAGCTT GAAGGAGCATCTCCTATGCG-3′; the underline is the restriction site of HindIII)

[0042] For the extraction of total RNA, the japonica rice variety Zhonghua 11 (or ZH11, a publicly popularized and applied rice variety cultivated by the Institute of Crop Science, Chinese Academy of Agricultural Sciences) was selected. RNA. The extraction reagent used TRIZOL reagent (purchased from Invitrogen Company), and the extraction method was operated according to the instructions of the above-mentioned TRIZOL reagent. Use reverse transcriptase M-MLV (purchased from TaKaRa Company) to synthesize cDNA by reverse transcription, according to the instruction manual of T...

Embodiment 2

[0067] The seedlings were cultivated by the soil culture method, and transplanted to the experimental field when the seedlings grew to the 4-leaf stage. Transplanting in three plants, the scale is 4 rows×12 beads, the spacing between plants and rows is 13.5×25cm, and the conventional fertilizer and water management is used. After the seedlings entered the tillering stage, the tiller angle was counted, and after heading, the tiller number was counted. Ear length and seed setting rate were counted in the later stage ( Figure 6 ).

[0068] Through the OsRUS1 overexpression vector ( figure 2 A) and RNAi vector ( figure 2 B) construction, using Agrobacterium-mediated transformation of rice embryogenic callus to obtain a large number of transgenic lines ( image 3 ), among the obtained two independent homozygous transgenic lines, the nucleic acid expression levels of the overexpressed transgenic lines were respectively increased by 19 times and 17 times compared with the wild...

Abstract

The invention discloses an OsRUS1 protein and an application of encoding genes thereof in control of tiller angle and tiller number of rice. The amino acid sequence of the OsRUS1 protein is shown as SEQ ID NO.1, and the amino acid sequence of the encoding genes of the OsRUS1 protein is shown as SEQ ID NO.2. The inventor finds that the OsRUS1 protein has a function of controlling the tiller angle and tiller number of rice, and according to overexpression, plant type looseness, tiller reduction and rice ear lengthening are caused. According to the OsRUS1 protein and the encoding genes thereof, the tillering mechanism or signal path of rice can be further researched; and rice varieties with loose or compact plant types can be obtained by inhibiting or promoting the expression quantity of the OsRUS1 protein.

Description

technical field [0001] The invention relates to the technical field of plant genetic engineering, in particular to the application of OsRUS1 protein and its coding gene in controlling rice tiller angle and tiller number. Background technique [0002] Rice is one of the most important food crops in the world, and more than half of the population depends on rice as a staple food. The Green Revolution in the 1960s greatly increased rice production and solved the then global food crisis. However, with the continuous growth of population and the sharp decrease of arable land, food security has become a global issue, so cultivating rice varieties with higher and more stable yields has become an important way to increase food security. Among the many factors affecting rice yield, the tiller angle and tiller number of rice are important agronomic traits that affect rice yield by affecting the effective use of sunlight and the effective panicle number of rice, and have always been v...
