Embodiment 1

[0029] 1. Materials and methods

[0030] The test materials were taken from the Xiaotangshan National Flower Engineering Center in Changping District, Beijing at the beginning of April 2011. figure 1 . The tender stems are used as explants to collect from the peony cultivar 'Coral Sunset' which is more than 3 years old.

[0031] Rinse the retrieved tender stems under running water, then soak them with detergent and 0.26% NaClO by mass for 20-30 minutes, then rinse them under running water for 30-40 minutes, absorb the water with absorbent paper, and place Go to the ultra-clean workbench for disinfection. Disinfect with 75% alcohol by volume for 30 seconds, rinse twice with sterile water, and then use 0.1% HgCl by mass 2 Disinfect for 10 minutes, rinse with sterile water for 4 times, cut the stem section into 0.1-0.3cm thin slices, and inoculate on the surface of the induction medium.

[0032] Inoculate the stem segments of 'Coral Sunset' on the starter medium based on thre...

Abstract

The invention belongs to the field of tissue culture of plants, and particularly provides a peony callus induction method which specifically comprises the following steps: sequentially inoculating the sterilized and disinfected peony stem into a start medium, an induction medium, an enrichment medium and a differential medium and performing conventional culture; and inoculating the obtained callus into an adventitious root induction medium and performing illumination culture or dark culture to obtain the tissue culture seedlings of peony. The peony callus induction method provided by the invention is simple to operate and low in cost, can be widely applied to multiple peony varieties, and effectively solves the problems that the traditional division propagation of peony cannot meet the needs of industrialized production and the underground tissue culture of peony is difficult. The invention also provides an application of the peony callus induction method in peony propagation.

Description

technical field [0001] The invention belongs to the field of plant tissue culture, and in particular relates to a callus induction method of peony. Background technique [0002] Peony (Peaonia lactiflora Pall.) is a perennial herbaceous flower of the Paeoniaceae genus Paeoniae. It has a long history of cultivation in my country, with a history of more than 3900 years. [0003] Peony is usually propagated mainly by ramets, once every 3-5 years, with a longer reproductive cycle and a lower reproductive coefficient. Seeding propagation method is relatively simple, but the reproduction cycle is long, usually 4-5 years before flowering, and the excellent traits of the female parent cannot be maintained, so it is not suitable for large-scale commercial production. Tissue culture can avoid the above disadvantages, and has the advantages of fast propagation speed and not limited by environmental conditions, so it can be promoted on a large scale. Using leaves and stems of plants a...
