Problems solved by technology

To date, there are no widely accepted clinical solutions for: crossing the "long gap" barrier so that fabricated structures can bridge nerve gaps longer than 2 to 3 cm; increasing the rate of nerve regeneration; or Improve the quality of nerve regeneration

Embodiment 1

[0106] Silicone tubing is biologically inert and has been used extensively in nerve regeneration research (Lundborg et al., 1982b; Williams et al., 1983). Lundborg showed that the rat sciatic nerve never regenerated through the empty silicone tube when the gap was longer than 15 mm. In contrast, we have shown that rat sciatic nerves can regenerate across at least 30 mm in empty silicone tubes when treated with the invention disclosed herein. The 30 mm gap was chosen because it is non-regenerating and thus represents the most difficult condition for nerve regeneration and is conserved across all species (Strauch et al., 2001). Furthermore, we have shown that rat sciatic nerves can regenerate across gaps of at least 50 mm in empty silicone tubes when treated with the invention disclosed herein. Furthermore, we have shown that the invention disclosed herein can also improve the outcome of the simplest type of nerve injury typified by sciatic nerve transection and immediate repai...

Abstract

The present disclosure describes the use of immunomodulators to promote nerve growth and regeneration, particularly in the case of neurological defects caused by trauma and disease. In particular, the present disclosure provides for the treatment of neurodeficit disorders using a CXCR4 antagonist, a STATS activator, and a nitric oxide enhancing agent, alone or in combination.

Description

[0001] priority statement [0002] This application claims the benefit of priority to U.S. Provisional Application Serial No. 62 / 871,552, filed July 8, 2019, the entire contents of which are hereby incorporated by reference. [0003] background 1. Technical field [0004] The present disclosure relates generally to the fields of medicine and neurobiology. More particularly, it relates to compositions and methods for treating nerve deficits and nerve damage. In particular, it relates to the treatment of these conditions using CXCR4 antagonists, STAT3 activators and nitric oxide enhancing agents alone or in combination. 2. Background technology [0005] Peripheral nerve injuries requiring surgical intervention occur in approximately 550,000 patients each year in the United States alone (published by Magellan Medical Technology Consultants, Inc., MN). This great clinical need has driven peripheral nerve regeneration research (Yannas et al., 2007). When damaged, the PNS has ...
