Embodiment 1

[0031] One, the preparation of irisaxanthin in kudzu flower

[0032] The specific operation steps of preparing irisaxanthin from kudzu flower are as follows:

[0033] (1) Raw material processing

[0034] The kudzu flower is dried, crushed and passed through a 60 sieve to obtain the kudzu flower powder.

[0035] (2) Gehua water extraction pretreatment

[0036] (2.1) Take 0.250g of kudzu flower powder, add deionized water according to the solid-to-liquid ratio of 1g: 60 mL, and ultrasonically extract at 50W / 45kHz for 1.5 hours;

[0037] (2.2) Centrifuge at 5000 rpm for 5 min to obtain the precipitate of kudzu flower powder;

[0038] (2.3) Shake and wash the precipitate with 10 mL of deionized water, centrifuge at 5000 rpm for 5 min, and remove the washing liquid; wash repeatedly 5 times;

[0039] (2.4) Dry the obtained precipitate below 60°C for 24 hours to obtain Pueraria flower powder pretreated by water extraction.

[0040] (3) Preparation of irisaxanthin

[0041] (3.1)...

Embodiment 2

[0055] The specific operation steps of preparing irisaxanthin from kudzu flower are as follows:

[0056] The difference between the method and embodiment 1 of kudzu flower water extraction pretreatment in this instance is:

[0057] In step (2), 12.5 mL of deionized water (solid-to-liquid ratio: 1 g:50 mL) was added to 0.250 g of Pueraria japonica, and the time for water extraction and ultrasound was 2 h. The rest of the operations and conditions were the same as in Example 1.

[0058] In step (3), the concentration of ethanol was 50%; the extract was filtered through a 0.22 μm filter membrane and freeze-dried, and the rest of the operations and conditions were the same as in Example 1 to obtain 6.16 mg of light yellow powder.

[0059] Purity and yield detection The same method as in Example 1 was used for content detection, and the results showed that the obtained irisaxanthin had a purity of 93.8% and a yield of 2.46%.

Embodiment 3

[0061] The specific operation steps of preparing irisaxanthin from kudzu flower are as follows:

[0062] The difference between the method and embodiment 1 of kudzu flower water extraction pretreatment in this instance is:

[0063] In step (2), add 4 mL of deionized water (solid-to-liquid ratio: 1 g:16 mL) to 0.250 g Pueraria japonica, and the rest of the operations and conditions are the same as in Example 1.

[0064] In step (3), the concentration of ethanol was 100%, and the ultrasonic extraction time was 60 minutes; after the extract was filtered through a 0.22 μm filter membrane and freeze-dried, the rest of the operations and conditions were the same as in Example 1, and 5.81 mg of light yellow powder was obtained.

[0065] Purity and yield detection The same method as in Example 1 was used for content detection, and the results showed that the obtained irisaxanthin had a purity of 93.2% and a yield of 2.32%.

Abstract

The invention relates to a method of preparing tectorigenin from flowers of kudzuvine. The method comprises the following steps: (1) carrying out aqueous extraction pre-treatment on flower powder of kudzuvine to obtain pre-treated flower powder of kudzuvine; and (2) carrying out ultrasonic flower powder of kudzuvine on the pre-treated flower powder of kudzuvine and then carrying out filtration bymeans of a filter membrane and freeze drying to obtain tectorigenin powder, wherein the content of tectorigenin in the tectorigenin powder is greater than 90%, and the yield is 2.17-2.7%. The purity of tectorigenin prepared from the flowers of kudzuvine is greater than 90% and the yield is about 2.7%. The high purity tectorigenin prepared by the method can serve as a food additive for producing health-care products for preventing tumors, diabetes and the like. Besides ethanol, no other organic reagents are used in an operating method, so that the method is an environment-friendly process method. The operating method provided by the invention is simple and relatively low in demand on equipment, expensive production facilities need not to be invested, the production cost is low, and the method is suitable for industrial batch production.

Description

technical field [0001] The invention belongs to the technical field of separation of plant natural products, and in particular relates to a method for preparing irisaxanthin from kudzu flowers. Background technique [0002] Irisxanthin is a specific isoflavone compound, mainly derived from Iridaceae plants such as Shegan and Gehua, and has a higher content in Pueraria (about 5-10% of the total flavonoids). Existing studies have shown that irisaxanthin has a good relieving effect on diabetic complications, and can also be used to treat a series of diseases such as cardiovascular and cerebrovascular diseases, and has great application potential in the fields of food additives and health products. [0003] At present, irisxanthin is mainly isolated from Iridaceae plants such as Shegan. The traditional method involves several steps of extraction with different organic solvents, batch concentration, and multiple separations on silica gel chromatography columns, and there are pro...
