Abstract
The invention discloses a functionality detection and evaluation method for liver disease NK (natural killer) cells, comprising the steps of (1) passing single cells through a flow cytometer, and using a combination of different anti-cytokine or NK cell surface receptor antibodies and cellular surface or intracellular specific markers to detect corresponding cytokine secretion or surface receptorlevels; (2) using a big data bio-statistical analytic technique to interpret the contribution, action and influence of each immune factor upon whole immunity, and finally NK cell overall immunity interpretation for the action of the immune factors upon immunity. The invention also provides a functionality detection kit for liver disease NK cells, comprising the surface molecular antibodies: CD56,NK-IFN-gamma, NK-TNF-alpha, NK-G2D, NK-p46, NK-p30, NK-p44, NK-KIRs, NK-LAIRs, NK-Perforin, NK-GrazymeB, NK-G2A, NK-PD-1, and NK-Tim3. The functionality detection and evaluation method and functionality detection kit herein can effectively detect overall functionality of NK cells.
Description
Technical field [0001] The invention belongs to the field of biomedicine and immunoassays, and relates to a method for detecting and evaluating the overall function of natural killer cells (NK) related to liver disease, and a kit for the method. Background technique [0002] Liver disease is a serious hazard to human health, and immune pathways are one of the most important strategies for liver disease diagnosis and treatment. [0003] my country is a major liver disease country. There are currently close to 200 million liver diseases, including hepatitis B, hepatitis C, alcoholic liver, fatty liver, drug-induced hepatitis, and autoimmune hepatitis; the trilogy of diseases from hepatitis-cirrhosis-liver cancer is a serious threat For human health, there are 600,000 deaths due to liver disease in my country every year, and the annual medical expenses related to liver disease are as high as 200 billion, causing serious economic and social burdens and exhaustion of medical resources. ...
