Embodiment Construction

[0028] The following describes embodiments of the apparatus and method of the present invention for analyzing a sample of cells without affecting the cells using digital holographic microscopy.

[0029] The embodiment shown in FIG. 1 includes a phase contrast microscope 1 . Said embodiment also includes a laser 2 emitting light at a wavelength of 633 nm, for example a He-Ne laser. The beam from the laser 2 is directed via a mirror 3 to a first beam splitter 4 which splits the beam into a target beam and a reference beam. The target beam passes through the spatial filter 5 and is directed to the second beam splitter 7 . The sample 8 is positioned such that the target beam reaches the beam dump arrangement 9 after passing through the sample. The reference beam passes through the spatial filter 6 and is reflected by the mirror 10 which redirects the reference beam towards the beam dump arrangement 9 . Such as figure 2 As shown, the reference beam 18 and the target beam 15 tra...

Abstract

A non-destructive method and device for analyzing a sample comprising transparent living and / or dead cells, by means of a digital holographic microscope, where the sample (8) is exposed to light from a laser (2). The light that travels through the cells in the sample will experience a difference in the optical path length compared to the surrounding media and the wave front that emerges from the cells will thus be phase shifted. This distortion can be detected in the digital hologram, which is reconstructed from the interference pattern detected by a digital sensor, such as a CCD or a CMOS, as phase differences or phase shifts and thereby creating a digital hologram. The phase shift of each element of the hologram is then used for analyzing the characteristics of the cells in the sample.

Description

technical field [0001] The present invention relates generally to the field of cell analysis. More specifically, the present invention relates to a non-destructive method and device for the analysis and characterization of samples containing live and / or dead cells in cell culture vessels by means of digital holographic microscopy. This can be used to measure the proliferation and survival of cells in a sample. Background technique [0002] To analyze nearly transparent cells, the cells are usually stained or made visible in some way. Because of the possibility of killing the specimen and the need to study the life processes of the specimen, the above methods are unsatisfactory in many respects. For example, when determining the number of cells in a cell culture vessel, the above methods are also time-consuming and therefore expensive. [0003] Therefore, there is currently a need for a new method for analyzing samples that does not affect the samples. [0004] Digital ho...
