An embodiment of the invention discloses a multipoint positioning method and device for a single-conductive-layer touch screen, wherein the multipoint positioning method comprises the following steps of: receiving a touch signal, and generating an original matrix which takes a minimal detection unit as a unit according to the triggering of the touch signal; respectively calculating a capacitance change quantity of each minimal detection unit in a touch signal induction region, and extracting the minimal detection unit of which the capacitance change quantity reaches a minimal calculation threshold value and is equal to a capacitance change quantity maximal value; dividing a calculation matrix corresponding to a minimal detection unit to be divided when the capacitance change quantity is more than the minimal detection unit to be divided under a preset condition; and calculating the position of the minimal detection unit with the capacitance change quantity maximal value in the original matrix, searching the minimal detection unit of which the capacitance change quantity reaches the minimal calculation threshold value and is equal to the capacitance change quantity maximal value from the calculation matrix, and calculating the position of the minimal detection unit in the calculation matrix.
